摘要
目的:探讨肝细胞癌(HCC)组织中ABCG2和Jagged1的表达状况,分析两者与肝癌临床病理学参数的相关性及临床意义。方法:分别应用免疫组化SP法和原位杂交的方法检测150例HCC、50例肝硬化和10例正常肝组织中ABCG2和Jag-ged1蛋白及mRNA的表达情况。结果:ABCG2蛋白在HCC、肝硬化和正常肝组织的阳性表达率分别为78.67%(118/150)﹑40.00%(20/50)和30.00%(3/10),差异有统计学意义,χ2=31.98,P<0.05。ABCG2mRNA在三种组织中的阳性表达率分别为69.33%(104/150)、36.00(18/50)和20.00%(2/10),差异有统计学意义,χ2=23.85,P<0.05。ABCG2蛋白在HCC组织中的表达与有无转移有关,χ2=10.93,P<0.05。Jagged1蛋白在HCC﹑肝硬化﹑正常肝组织中的阳性表达率分别为71.33%(107/150)、52.00%(26/50)和20.00%(2/10),差异有统计学意义,χ2=15.07,P<0.05。Jagged1mRNA在三种组织中的阳性表达率分别为60.67%(91/150)、36.00%(18/50)和20.00%(2/10)差异有统计学意义,χ2=13.71,P<0.05。Jagged1蛋白表达与年龄(χ2=7.94,P<0.05)、组织学分级(χ2=14.79,P<0.05)及HBsAg(χ2=4.83,P<0.05)有关。ABCG2蛋白的表达与Jagged1蛋白表达呈正相关,r=0.458,P<0.05。结论:ABCG2和Jagged1可能成为判断HCC预后和分子生物学行为的重要指标。
OBJECTIVE: To explore the expression of ABCG2 and Jaggedl in human hepatocellular carcinoma, and the re- lationship between the expressions and clinicopathological characters. METHODS: The expressions of ABCG2 protein and Jag- gedl protein in 150 HCC tissues, 50 cirrhosis tissues and 10 normal liver tissues were detected by immunohistochemical method, and the expressions of ABCG2 mRNA and Jaggedl mRNA were assessed by in situ hybridization. RESULTS: The expression rates of ABCG2 protein in cancer tissues, cirrhosis tissues and normal liver tissues were 78.67 % (118/150), 40.00 % (20/50) and 30.00%(3/10) with significant difference (χ2= 31.98, P〈0.05), while those of ABCG2 mRNA were 69.33% (104/150), 36.00% (18/50) and 20.00% (2/10) with significantly difference (χ2= 23.85,P〈0.05). There was correlation of ABCG2 pro- tein expression with the metastasis in HCC (χ2= 10.93 ,P〈0. 05). The expression rates of Jaggedl protein in cancer tissues, cir- rhosis tissues and normal liver tissues were 71.33 % (107/150), 52.00 % (26/50) and 20.00 % (2/10) with significant difference (χ2 = 15.07 ,P〈0.05) ,while those of Jaggedl mRNA were 60.67 % (91/150) ,36.00% (18/50) and 20.00% (2/10) with sig- nificantly difference (χ2 = 13.71 ,P〈0.05). The Jagged1 protein expression was correlated with the age (χ2 = 7.94,P〈0.05), Edmondson grade (χ2 = 14.79,P〈0.05) and HBsAg (χ2 = 4.83,P〈0.05). Besides, ABCG2 expression was positively correla- ted with Jagged1 expression in HCC(r=0. 458,P〈0. 05). CONCLUSION: ABCG2 and Jagged-1 may be important prognostic indicators and potentially useful for assessing the biological behavior of HCC.
