摘要
目的探讨人宫颈鳞状上皮细胞癌(简称鳞癌)组织中电导钾离子通道(IKCal)mRNA及其蛋白产物与正常宫颈上皮组织中钙激活性的差异。方法采用免疫组织化学法及RT-PCR法分别检测30例不同分化程度的宫颈鳞癌组织和18例正常宫颈上皮组织中IKCalmRNA及其蛋白产物的表达情况。结果(1)IKCalmRNA阳性表达率15例子宫颈鳞癌组织中分别为73.3%,表达水平为(1.2±n5),18例正常宫颈上皮组织中分别为11.1%,表达水平为(0.9±0.2),差异均有统计学意义;(2)30例宫颈鳞癌组织细胞核及细胞膜上IKCal蛋白产物均呈阳性表达;正常宫颈上皮组织中仅1例在鳞状上皮部位细胞膜部位有较弱表达,差异有统计学意义;(3)宫颈鳞癌组织中IKCal蛋白产物的表达与癌细胞分化程度呈正相关。结论IKCal的高表扶与宫颈鳞痛的发牛和发展密切相关.
Objective To compare the expression level of intermediate-conductance- Ca2+ -activated K + channels (IKCal) mRNA and proteins in the tissue of cervical cancer with that of normal cervical tissue. Methods Reverse transcriptase polymerase chain reaction (RT-PCR) and immunohistochemical assay were used to quantify mRNA and pro- tein expression in cervical cancer tissue and normal cervical tissue, and to examine the relationships between IKCal and the risk of cervical cancer. Results In the 15 specimens of cervical cancer, the expression rate of IKCal mRNA was 73.3%, and the expression level was 1.1 ~ 0.5. In the 18 specimens of normal cervix, the expression rate of IKCal mR- NA was 11.1%, and the expression level was 0.9 ± 0.2. The expression rate and level of IKCal mRNA in cervical cancer were higher than those of normal cervix. All the specimens of cervical cancer were expressing the IKCal protein in the nu- cleus, compared with only 1 case expressing the IKCal antibody in normal cervix. The expression rate of IKCal protein in cervical cancer was higher than that of normal cervix. The expression of IKCal protein in cervical cancer was related to the degree of differentiation of the cancer cells. Conclusion Higher expression level of IKCal mRNA and proteins was noted in cervical cancer. Over-expression of IKCal may be related to the occurrence and development of cervical cancer.
