摘要
目的:比较虎杖药材中白藜芦醇及其苷的两种含量测定方法。方法:采用高效液相色谱(high performance liquid chromatography,HPLC)串联紫外-荧光检测器法同时测定虎杖乙醇提取物中白藜芦醇及其苷的含量。C18色谱柱,乙腈-水为流动相,紫外检测波长λ=290 nm,荧光波长λEx/λEm=334/404。结果:两种方法线性范围、回收率、精密度等试验的比较结果基本一致,但荧光检测法灵敏度相对较高。结论:两种检测方法均能准确测定虎杖中白藜芦醇及其苷的含量。
Objective: To compare the two methods for content determination of resveratrol its glucoside. Methods: Reverse phase high performance liquid chromatography(RP-HPLC) with Uv and Flu detector in tandem was used to measure the contents of resveratrol and its glucoside in Polygonum Cuspidatum extrae. A Shimadzu C18 column(250 mm×4.5 mm, 5 μm) was used for chromatographic separation. The mobile phase was a two-element system composed of acetonitrile-water for gradient elution at a flow rate of 1.0 mL/min. The ultraviolet and fluorescence detector wcre employed for detection with their wavelength of 290 nm and λFx/λFm=334/404, respectively. Results: By comparing the results of accuracy, precision, linearity, etc of the two methods, no significance was found. But the main difference between them was that the sensitive for Flu detector was about teen times higher than that of Uv detector. Conclusion: These two methods were all fit to the content determination of resveratrol and its glueoside in Polygonum cuspidatum extract.
