摘要
目的通过白虎汤对肺炎双球菌肺炎大鼠自由基、前列腺素代谢及C-反应蛋白(CRP)和铜蓝蛋白(CP)含量的影响,探讨白虎汤的抗炎作用。方法用气管插管法制作肺炎大鼠模型,分为正常组、模型组、白虎汤低剂量组、白虎汤高剂量组、头孢氨苄组。正常组与模型组处理为灌胃生理盐水,白虎汤治疗低剂量组、高剂量组及头孢氨苄组分别灌胃白虎汤(9 g.kg-1.d-1,18 g.kg-1.d-1)、头孢氨苄(2.7 g.kg-1.d-1)。观察肺脏组织的病理改变及各组血清和小肠组织中SOD活性、MDA、NO、TXB2、6-Kete-PGFla及血清的C反应蛋白的变化。结果肺炎大鼠肺组织损伤明显,造模后大鼠血清中的NO、MDA、TXB升高,SOD活性、6-Kete-PGFla降低,CPR和CP升高,给予低剂量、高剂量白虎汤治疗后,血清及小肠组织中SOD活性、6-Kete-PGFla含量升高,MDA、NO、TXB2含量下降,CPR和CP降低,白虎汤低剂量组对于小肠组织中的SOD活性作用不明显,与白虎汤高剂量组及头孢氨苄组相比,有显著差异(P<0.05),而白虎汤高剂量组与头孢氨苄组在改善SOD活性、MDA、NO、TXB2、6-Kete-PGFla及CPR和CP方面,无明显差异(P>0.05)。结论白虎汤具有较好的抗炎作用,能够拮抗自由基损伤及调节前列腺素代谢,降低CPR和CP,保护肺组织免受损伤。
Objective To explore the anti - inflammatory effects of Bai Hu decoction, by studying the effects of pneumococcal pneumonia in rats free radicals, prostaglandin metabolism and the impact of C - reactive protein (CRP) and ceruloplasmin (CP) content. Methods We first established pneumonia rat model by tracheal intubation. Rats were divided into 5 groups: normal group, model group, low dose group of Bai Hu decoction, high dose group of Bai Hu decoction, cephalexin group. Low dose group, high dose group of Bai Hu decoction and the cepbalexin group were intragastric administered with dose 9 g kg- 1 . d- l, 18 g kg-l d-1 and 2.7 g kg-1 d-l, normal control group and model group were given same volume of saline. Then ob- served the pathological changes of lung tissue and the activity of SOD, MDA, NO and TXB2,6 -Kete -PGFla and serum C -re- active protein changes in serum and intestinal tissue. Results Lung tissue injury was obviously in pneumonia rat, and NO, MDA and TXB increased, the activity of SOD and 6 - Kete - PGFla lowered, CPR and CP increased significantly in rat serum after modeling. After treatment, SOD activity and 6 - Kete - PGFla increased, MDA, NO, TXB2 , CPR and CP decreased in serum and intestinal tissue of low dose group and high dose group of Bai Hu decoction. Change of SOD activity in the intestinal tissue was not obvious in the low close group of Bai Hu decoction, compared with high - dose group of Bai Hu decoction and the cepha- lexin group ( P 〈 0.05 ). While there was no significant difference ( P 〉 0.05 ) between the high dose group and the cephalexin group of Bai Hu decoction, to improve the activity of SOD, MDA, NO, TXB2, 6 - Kete - PGFla, and CPR and CP. Conclusion Bai Hu decoction has good anti - inflammatory effect, antagonizes the free radical damage and regulates prostaglandin metabolism, reduces the CPR and the CP, and protects lung tissue from damage.
