淫羊藿总黄酮抑制单核细胞分泌M-CSF对人工关节松动的影响

摘要 目的探讨淫羊藿总黄酮对人工关节无菌性松动的防治作用。方法分离人外周血单核细胞体外培养,加入超高分子聚乙烯、金属微粒及不同浓度的淫羊藿总黄酮培养后,放免法测定各组细胞上清液中巨噬细胞集落刺激因子(M-CSF)的含量。结果超高分子聚乙烯、金属微粒组的含量明显高于对照组,淫羊藿总黄酮组的含量明显低于超高分子聚乙烯、金属微粒组而不同浓度组淫羊藿总黄酮之间的差异无统计学意义。结论超高分子聚乙烯、金属微粒可刺激单核细胞过量分泌M-CSF,淫羊藿总黄酮能有效地抑制单核细胞的过量分泌,可能在防治人工关节松动中起到作用。 Objective To explore the possibility of TFE contributing to aseptic loosening of orthopedic implants. Methods 36 samples of peripheral blood were obtained from Femoral neck fracture donors. Monocytes (MOs) were isolated and assigned to 8 groups (n =36 each) for culture. Group A was treated with MOs alone; group B with MOs 4 UHMWPE ; group C with MOs + Metal ions ; group D with MOs + UHMWPE + Metal ions ; group E with MOs + UHMWPE + Metal ions + TFE(5ug/ml) ; group F with MOs + UHMWPE + Metal ions + TFE(10ug/ml) ; group G with MOs + UHMWPE + Metal ions + TFE (20ug/ml) ; group H with MOs + UHMWPE + Metal ions + TFE (40ug/ml). The ceils were co-cultured for 48 hours and the supernatant was tested for M-CSF content with radioimmunoassay. Results The levels of M-CSF were much higher in group B, C, D than in group A. There was no significant difference in M-CSF levels between group B and group C ( P 〉 0. 05 ). Conclusions UHMWPE, Metal ions effectively stimulates MOs to secrete M-CSF. It seems that TFE can be a notential theraneutic agent for the prevention and treatment of aseptic loosening of joint prosthesis.
