摘要
为探讨离子注入对石蒜属植物种子生物化学特性的影响,采用不同剂量的钛离子注入3种石蒜属植物种子,研究注入后钛离子对种子电解质外渗率(REC)、丙二醛(MDA)、超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)和发芽率的影响。结果表明:随着钛离子注入剂量的增加,3种石蒜属植物种子相对电解质外渗率不断上升;MDA含量先升后降,随后又小幅回升;SOD、POD活性先升后降,但峰值因离子注入剂量和植物不同而异;石蒜和换锦花种子CAT活性先升后降,中国石蒜种子CAT活性呈"升-降-升"的变化趋势;石蒜种子发芽率曲线呈"马鞍型",中国石蒜和换锦花种子发芽率呈"双马鞍型"曲线,能量为30keV的Ti+诱变石蒜种子的适宜剂量为25×1016ions.cm-2,适于中国石蒜和换锦花种子诱变的注入剂量分别是50×1016ions.cm-2和500×1016ions.cm-2。一定剂量的钛离子注入能激活细胞合成自由基清除酶的能力,3种石蒜属植物的种子发芽率、REC、MDA、SOD、POD以及CAT间存在不同程度的相关性,酶类物质通过相互协调作用减低自由基造成的伤害。
In order to investigate the effects of ion implantation on biological and chracteristics of seeds from lycoris, different doses of Titanium ion were injected into the seeds of three species of Lycoris. The effects on relative electric conductivity( REC), malondiadehyde (MDA) content,superoxide dismutase (SOD) activity, peroxidase (POD) activity, catalase (CAT) activity and the percentage of seeds germination were studied. The results indicated: along with doses of Titanium ion increasing, REC increased, MDA content rising first, and then falling and rising again, POD increased first and then decreased, but the maximum values for three plant species was vary with different species. CAT activity in seeds of L. radiata and L. sprengeri first increased and then decreased, it was rising first, and then falling and rising again in seeds of L. chinese. Germination percentage of L. radiata alter in " saddle" line, but " double saddle" line appeared in germination percentage of L. chinese and L. sprengeri with dosing. The dose of 25 ×10^16ions·cm^-2 might be optimum for seed mutagenesis of L. radiata. The dose of 50× 10^16ions·cm^-2 and 500 ×10^16ions·cm^-2might be optimum for seed mutagenesis of L. chinese and L. sprengeri. It suggested that some certain dose of Titanium ion beam implanting can activate the ability of enzyme scavenging free radical. Correlation analysis showed that germination percentage, REC, MDA, SOD, POD, CAT were correlated with each other, the reaction between enzyme could defense free radical injury.
