杜仲翅果桃叶珊瑚苷高效液相色谱分析方法的建立

摘要 为建立检测杜仲翅果桃叶珊瑚苷含量的HPLC法,以杜仲翅果为原料,采用溶剂浸提法,用85%乙醇在提取温度50℃、料液比1∶20、提取时间30 min的条件下提取2次,浓缩提取液至无醇味,综合分析检测波长、流动相、流速、检测温度4个因素对HPLC图谱的影响。结果表明:在C18色谱柱(150 mm×4.6 mm,5μm)、检测波长208 nm、柱温20℃、流速0.8 mL/min的条件下,用1%~10%的乙腈梯度洗脱30 min,杜仲翅果桃叶珊瑚苷含量在样品进样量0.30~1.50μg与峰面积呈良好的线性关系,R2=0.999 6,平均回收率为84.3%,相对标准差为1.97%(n=5)。该方法可准确检测杜仲翅果乙醇提取物中桃叶珊瑚苷含量,具有样品处理简单、稳定性强、重现性好、精密度高的优点。 In order to detect the content of aucubin in samara of Eucommia ulmoides by high performance liquid chromatography(HPLC), Eucommia ulmoides samara was taken as raw materials, 85 percent ethanol was used at the condition of temperature 50 ℃, liquid ratio 1 : 20, extraction for 2 times and 30 rain for each time to extract aucubin till there is no alcohol taste in the concentrated extracts. The purpose of this research is to develop a set of useful HPLC analysis approach of Eucommia ulmoides aucubin from the detection wave length, mobile phase, flow velocity and temperature. The results showed that optimal separation was achieved on a Cl8 (150 mm×4.6 mm, 5 μm) column, whenits mobile phase ranged from 1 to 10 percent acetonitrile, gradient elution, and its flow rate was 0.8 mL/min, its column temperature was 20 ℃ and its wavelength was set at 208 nm. The calibration curve was linear within the range of 0.30-1.50 μg, R2=0.999 6, the average recovery for aucubin was 84.3%, RSD was 1.97%(n=5). This analysis method could be used to detect the content ofEucommia ulmoides aucubin with simple samples, stability, good repeatability and high precision.
