摘要
目的:观察离心运动致骨骼肌损伤后大鼠血清和骨骼肌甘氨酸、苏氨酸、精氨酸和赖氨酸含量的变化,探讨上述4种氨基酸在骨骼肌损伤和修复过程中的变化规律。方法:成年健康雄性SD大鼠在一次性90 min持续下坡跑运动(速度16 m/min,坡度-16°)后,分别在运动后0 h、6 h、12 h、1 d、2 d、3 d、1 w、2 w处死,取大鼠右侧腓肠肌,HE染色进行组织学观察,高效液相色谱法测试血清与骨骼肌中甘氨酸、苏氨酸、精氨酸和赖氨酸含量。结果:(1)HE染色结果显示,运动后6 h骨骼肌细胞有肿胀的趋势,2 d发现炎性细胞浸润,3 d炎性细胞最多,损伤程度最为严重,1 w偶见炎性细胞,2 w后肌纤维形态结构基本恢复正常。(2)运动后0 h血清4种氨基酸含量与运动前相比均显著下降(P<0.05,其中精氨酸为P<0.01),之后4种氨基酸含量出现不同程度上升或下降。2 w时血清赖氨酸水平仍显著低于运动前水平(P<0.05)。运动后0 h骨骼肌甘氨酸和赖氨酸含量出现上升趋势,运动后2 d和3 d赖氨酸含量显著高于运动前水平(P<0.05),2 w时与运动前水平相比无显著性差异。运动后0h骨骼肌苏氨酸和精氨酸含量均显著低于运动前水平(P<0.05),苏氨酸在运动后1 w、精氨酸在运动后2 d均显著低于运动前水平(P<0.05)。(3)血清精氨酸和甘氨酸,骨骼肌精氨酸和苏氨酸含量显著正相关(P<0.01)。血清甘氨酸和骨骼肌甘氨酸显著负相关(P<0.01),但与骨骼肌精氨酸显著正相关(P<0.05)。血清赖氨酸和骨骼肌苏氨酸、精氨酸显著正相关(P<0.01,P<0.05),血清精氨酸与骨骼肌精氨酸之间正相关(P<0.05)。结论:离心运动后大鼠血清和骨骼肌甘氨酸、苏氨酸、精氨酸和赖氨酸含量变化不同,但有不同程度的相关。血清赖氨酸含量在2 w时仍未恢复到运动前水平,提示骨骼肌修复期机体对赖氨酸可能具有更大需求。
Objective The purpose of this study is to investigate the change in the contents of glycine,threonine,arginine,and lysine in serum and skeletal muscle of rats after eccentric exercise. Methods Healthy adult male SD rats were sacrificed at 0 hour,6 hours, 12 hours, 1 day,2 days,3 days, 1 week, and 2 weeks after single bout of 90-min downhill running on treadmill at the speed of 16m/rainand slope of -16°. Right gastrocnemius was sampled and stained with HE to investigate its morphological profile,and the contents of glycine,threonine,arginine and lysine in skeletal muscle and serum were determined by HPLC. Results (1) Myofibers tended to be swollen at 6 hours after exercise, inflammatory ceils in myofibers appeared at 2 days after exercise and reached the maximum at 3 days after exercise. Inflammatory cells were scarce and morphology of myofibers became normal 1 week after exercise. (2) Serum glycine,threonine,and lysine concentrations lowered significantly immediately after exercise (P 〈 0.05) ,especially arginine (P 〈 0.01). They returned to baseline 2 weeks after exercise,except lysine. The content of glycine and lysine tended to increase immediately after exercise,and lysine increased significantly 2 and 3 days after exercise (P 〈 0.05) and became normal 2 weeks after exercise. The content of threonine and arginine in muscle decreased significantly immediately after exercise (P 〈 0.05) ,threonine 1 week after exercise,and arginine 2 days after exercise remained lower than before exercise (P 〈 0.05). (3) There was significant positive correlation between serum arginine and glycine, and skeletal muscle arginine and threonine (P 〈 0.01).
