摘要
采用响应面法优化趋磁细菌AMB-1产磁小体的条件。采用Plackett-Burman法对6个单因素进行筛选,再用最陡爬坡实验及Box-Behnken实验对以上结果进一步优化,利用SAS软件进行数据处理。Plackett-Burman实验结果表明奎尼酸铁添加量、培养温度和pH对趋磁细菌生长及磁小体产量的影响较大。优化结果:奎尼酸铁添加量为2.7μmol,培养温度为32℃,培养基初始pH为6.8。在此条件下,趋磁细菌OD600达到0.980,Cmag值为1.782。与基础培养基相比,菌种量和磁小体形成量分别提高了14.9%和16.5%。
Response surface method was adopted to optimize the conditions for the magnetosome production by magnetotacitic bacterium AMB-1. Plackett-Burman method was used to screen six single-factors. Then the steepest ascent and Box-Behnken experiments were used for further optimization, and data processing was carried out with SAS soft-ware. The results of Plackett-Burman experiment indicated that the addition amount of ferric quinate, the incubation temperature and pH affected fairly great on the production of magnetosomes and the growth of magnetotaeitic bacterium AMB-1. The optimized results were ferric quinate at 2.7 μmol, the incubation temperature at 32℃ and pH at 6.8. Therefore, under such conditions, the OD600 of magnetotacitic bacterium strain AMB-1 was 0.980 A, and the value of Cmag was 1. 782, the strain amount and the formation amount of magnetosome increased by 14-. 9% and 16.5% respectively as compared with basic culture medium.
