摘要
目的:利用高效液相色谱法—蒸发光散射检测器测定海南霉素的纯度。方法:采用苯基色谱柱(250mm×4.6,5μm),以甲醇-水-三氟乙酸(90∶10∶0.05)为流动相;流速为1.0mL/min;用蒸发光散射检测器(高温型,漂移管温度110℃,载气流量为2.5L/min)进行检测。检测结果显示海南霉素在0.5~2mg/mL的浓度范围内峰面积与浓度呈线性相关系,检测限和定量限分别为0.025mg/mL和0.05mg/mL。结论:采用高效液相-蒸发光散射检测法检测海南霉素纯度方便、快速、准确,可为进一步制定海南霉素质量标准提供依据。
Objective: To determine the purity of hainanmyein by HPLC-ELSD. Methods: Chromatography conditions: SB-Phenyl column with methanol-water-trifluoroacetic (90 : 10 : 0.05) as the mobile phase, flow rate 1. 0mL/min, an evaporative light-scattering detector (ELSD) Model2000E(Alltech , USA ) was used. Its parameters were set as follows: drift tube temperature at 110, and the flow rate of carrier gas at 2.5 L · min^-1. The calibration curve was good linear relationship in the range of 0. 5 -- 2. 0mg/mL ( r = 0. 996). Limit of detect and limit of quantification is 0. 025mg/mL and 0.05 mg/mL respectively. Conclution: The method used to determine the purity of hainanmycin by HPLC-ELSD is simple, rapid and accurate . it will help for establishment quality criterion of hainanmycin.
