养脾阴法对早期帕金森病大鼠模型行为学、纹状体多巴胺及其代谢产物的影响 被引量:7

Influence of Nourishing Spleen Yin Method on the Behavior Striatal Dopamine and Its Metabolites in Rat Model of Early Parkinson’s Disease

摘要 目的:探讨养脾阴法对早期帕金森病大鼠模型行为学及纹状体多巴胺及其代谢产物的影响。方法:采用小剂量(4μg/4μL)6-羟多巴胺单侧内侧前脑束注射法制备早期帕金森病大鼠模型,2周后据阿朴吗啡诱导的不自主运动,筛选造模成功大鼠,随机分为模型组、养脾阴组,另设假手术对照组,灌胃相应药物共8周。记录阿朴吗啡诱导的大鼠向健侧旋转的圈数,高效液相色谱法测定大鼠纹状体多巴胺(DA)、二羟苯乙酸(DOPAC)及高香草酸(HVA)的含量。结果:术后第4周、第8周养脾阴组大鼠向健侧旋转的圈数与模型组比较明显减少(P<0.01)。术后第8周模型组大鼠纹状体DA、DOPAC、HVA含量与假手术组比较显著降低(P<0.05),养脾阴组纹状体DA、DOPAC、HVA含量与模型组比较明显升高(P<0.05或P<0.01),但较假手术组降低(P<0.05或P<0.01)。结论:养脾阴法可能对多巴胺能神经元具有神经保护作用。 Objectives:To investigate the influence of nourishing spleen yin method on the behavior, striatal dopamine and its metabolites in rat model of early Parkinson's disease. Methods : Rat model with early Parkinson's disease was established by unilateral injection of low dose(4μg/4 μL) of 6 - hydroxydopamine into medial forebrain bundle. Two weeks later, the successful modeling rats were screened out according to the involuntary movements induced by apomorphine, and the rats were randomly divided into model group, nourishing spleen yin group and sham -operation control group. The appropriate drugs were administered by garage for a total of eight weeks. The number of turns of apomorphine induced contralateral rotation was recorded. High performance liquid chromatography method was used for the determination of con- tents of dopamine ( DA), dihydroxyphenylacetic acid ( DOPAC ) and homovanillic acid (HVA) in rat striatum. Results : Compared with model group, nourishing spleen Yin method significantly decreased the number 9f contralateral turns at 4 and 8 weeks post -operation(P 〈 0.01 ). In 8 weeks post -operation, the striatal contents of DA, DOPAC and HVA in model group were significantly decreased compared with that in sham operation group(P 〈 0.05 ). The striatum contents of DA, DOPAC and HVA in nourishing spleen Yin group were significantly higher than that in the model group( P 〈 0.05 or P 〈 0.01 ) ,but lower than that in the sham - operation group( P 〈 0.05 Or P 〈 0. 01 ). Conclusion: Nourishing spleen yin method may have neuroprotective effects on dopaminergic neurons.
