巴戟天及不同盐分炮制品中糖类成分HPLC-ELSD指纹图谱研究 被引量:9

HPLC-ELSD Fingerprint of Carbohydrate Constituents in Morindae Officinalis Radix and its Salt Processed Products

摘要 目的:建立巴戟天及不同盐分炮制品中糖类成分HPLC-ELSD指纹图谱分析方法,为确定盐制巴戟天饮片在炮制过程中的用盐比例及其质量标准的制定提供依据。方法:采用HPLC,色谱柱为Agilent ZORBAX Carbohyrate(4.6 mm×250mm,5μm),流动相为乙腈-甲醇-水,流速1.2 mL.min-1,梯度洗脱,柱温35℃,以蒸发光散射检测器(ELSD)检测,漂移管温度为105℃,增益值为1,氮气流量2.0 L.min-1;采用国家药典委员会推荐的《中药色谱指纹图谱相似度评价系统》(2004年A版)进行评价。结果:以耐斯糖峰为参照峰,18批样品标定了共有峰16个,各样品相似度均>0.90。结论:该方法简单、准确、重复性好,可用于巴戟天及其盐制品糖类成分的质量控制。 Objective: To establish the HPLC-ELSD fingerprint analysis method of carbohydrate constituents in Morindae Officinalis Radix and its salt processed products and provide the basis for the amount of salt proportions when being processed and quality standard. Method: HPLC analysis was performed on a Agilent ZORBAX carbohyrate analysis column (4.6 mmx 250 mm, 5 m) with a mixture of methanol and acetonitrile and water as mobile phase in gradient mode. The drift tube temperature was at 105 ℃ , nitrogen flow was 2.0 L -1 mln , gain value was 1, column temperature was at 35 ℃ and flow rate was 1.0 mL min-1 The chromatogram fingerprint was evaluated by the similarity evaluation software published by committee of pharmacopeia. Result: Sixteen co-possessing peaks were selected as the fingerprint peaks of Morindae Officinalis Radix by taking nystose peak as the referential peak. The fingerprint similarity value of eighteen batches was above 0.90 compared with system generation spectrum. Conclusion: The method is simple, accurate and reproducible; it can be used for quality control of carbohydrate components in Morindae Officinalis Radix and its salt processed products.
