摘要
目的:探讨电针联合脂肪源性干细胞(ADSC)移植对大鼠缺血/再灌注损伤后海马区胰岛素样生长因子1(IGF-1)及其受体(IGF-1R)表达的影响。方法:成年SD大鼠随机分为正常组、模型组、电针组、ADSC组、电针加ADSC组。NSS法行神经功能损害评分,Western bolt法及实时荧光定量PCR分别检测海马区IGF-1、IGF-1R蛋白和mRNA表达。结果:电针加ADSC组海马区PKH-26标记的细胞个数高于ADSC组。电针加ADSC组较电针组、ADSC组NSS评分显著降低,而海马区IGF-1和IGF-1R蛋白和mRNA表达显著增加。结论:电针加ADSC联合治疗协同促进脑缺血大鼠神经功能恢复,其机制可能与电针上调海马区IGF-1和IGF-1R的表达、改善干细胞生存内环境、促进移植的ADSC迁移和存活有关。
Objective:To investigate the effect of electroacupuncture(EA) combined with adipose-derived stem cell(ADSC) transplanted on IGF-1 and IGF-1R in hippocampus after cerebral ischemia-reperfusion in rats. Methods:Adult SD rats were randomly divided into normal group,model group,EA group,ADSC group and EA plus ADSC group.Neurological function score was observed by NSS.The expressions of IGF-1 and IGF-1R in hippocampus were observed by using Western bolt and quantitative real-time RT-PCR. Results:The number of PKH-26 labeled ADSC in EA plus ADSC group was higher than that in ADSC group.Compared with EA group and ADSC group,the protein and mRNA of IGF-1 and IGF-1R were increased in EA plus ADSC group.However,the NSS was significantly decreased. Conclusion:The combined treatment of electroacupuncture plus ADSC promotes functional outcome significantly and the efficacy is better than either of the single treatment,which may be related to EA upregulating IGF-1 and IGF-1R expressions in hippocampus,and promoting ADSC survival and migration.
