摘要
目的探讨hCG联合免疫球蛋白在免疫性流产的治疗中是否具有协同作用。方法 CBA/J雌性小鼠按体重编号后按随机数字表法随机分为4组,即自然流产模型组(CBA/J×DBA/2)、hCG用药组(CBA/J×DBA/2,hCG 35U/kg,肌注,隔天一次)、hCG联合免疫球蛋白组(CBA/J×DBA/2,hCG 35U/kg,肌注,隔天一次,免疫球蛋白,40mg/kg,肌注,孕第一天)。正常妊娠模型组(CAB/J×BALB/c)。孕第14天处死孕鼠,计算各组胚胎吸收率,留取蜕膜并进行组织匀浆,ELISA方法测定蜕膜匀浆液中TNF-α、IFN-γ、IL-4、IL-10的含量。结果自然流产组的胚胎吸收率高于其它三组,差异有统计学意义(P<0.05),hCG组的胚胎吸收率高于联合用药组,差异有统计学意义(P>0.05)。与正常妊娠组比较,自然流产模型组TNF-α、IFN-γ的水平明显高于前者(P<0.05),而IL-4及IL-10的水平低于前者,差异均有统计学意义(P<0.05)。hCG组和联合用药组TNF-α及IFN-γ水平明显低于自然流产组(P<0.05),而IL-4及IL-10高于自然流产组,联合用药组及正常妊娠组细胞因子水平差异无统计学意义(P<0.05)。结论 hCG联合免疫球蛋白通过调节母胎界面Th1/Th2型细胞因子水平来促进妊娠成功,二者具有协同作用。
Objective: To investgate the effect of the combine therapy of hCG puls human immunoglobin G on the maternal-fetal interface Th1/Th2 type cytokines of recurrent abortion model,and investgate the therapy′s effect on pregnancy′s outcome.Methods: Erect three abortion model CBA/J×DBA/2,and erect normal pregnancy model CBA/J×BALB/C.Then the abortion model randomly divided into 3 groups(5 mice in each group),abortion model group,injection of hCG group(35IU/kg,im,qod),injection of hCG(35IU/kg,im,qod) and human immunoglobin G(40mg/kg,im,st,only on the first day of pregnancy)group.On the 14th day after pregnancy,they were sacrificed by chloralic hydras.And observe the fetal resorption rate(%),leave a mount of decidua,which mingle with the RIPA and PMSF,then grind them fully in a glass container.Centrifuge and leave the supernatant.Evaluate the TNF-α,IFN-γ,IL-4,IL-10′s level of the supernatants by ELISA.Stastistical analysis was performed by one-factor analysis of variance and T-test.Results:Compared with the abortion model group,the combination of hCG and human immunoglobin G increase Th2 type cytokines(IL-4,IL-10,P0.05),and decrease Th1 type cytokines(TNF-α,IFN-γ,P0.05),and statistically reduced fetal absorption rate of abortion model(P0.05).Conclusion:The combination of hCG and immunoglobin G can regulate local maternal-fetal interface Th1/Th2 type cytokines and results in Th2 immune bias,then promote pregnancy outcome.
