摘要
目的建立灵芝子实体和孢子粉中3种灵芝酸类成分的HPLC测定方法 ,为全面评价灵芝药材和孢子粉的质量提供依据。方法采用HPLC方法测定,Diamonsi(l钻石)C8色谱柱(250mm×4.6mm,5μm),流动相乙腈-0.03%磷酸(28:72),体积流量1.2mL/min,检测波长250nm;柱温35℃。结果灵芝酸C2、灵芝酸G、灵芝酸A分别在0.40~10.06μg(r=0.99999)、0.40~10.00μg(r=0.99999)、0.40~10.00μg(r=0.99999)线性关系良好,平均回收率分别为101.04%、99.39%、101.22%,RSD值分别为1.64%、1.86%、2.20%(n=6);灵芝子实体中3种灵芝酸的质量分数分别在0.06~0.29mg/g、0.12~0.37mg/g、0.19~0.41mg/g,灵芝孢子粉供试品中3种灵芝酸的质量分数分别在0~0.01mg/g、0、0~0.03mg/g。结论本方法简便,稳定,可用于灵芝三萜类成分C2、灵芝酸G、灵芝酸A的定量分析。
Objective To provide the scientific evidence for overall quality evaluation of Ganoderma,a trial to develop HPLC method for quantitative analysis of three ganoderic acids in Ganoderma was carried out.Methods The HPLC method was performed on Diamonsil C 8 column (250mm×4.6mm,5μm),with acetonitrile-0.03% phospholic acid (28:72) as the mobile phase;The flow rate was 1.2mL/min;UV detection wavelength was 250nm;The column temperature was 35 ℃.Results The liner ranges of ganoderic acid C 2,ganoderic acid G and ganoderic acid A were 0.40—10.06μg (r=0.999 99),0.40—10.00 μg (r=0.999 99) and 0.40—10.00 μg (r=0.999 99);The average recoveries were 101.04%,99.39% and 101.22%,with RSD values of 1.64%,1.86%,and 2.20% (n=6),respectively.The content ranges of three ganoderic acids in sporophore were 0.06—0.29 mg/g,0.12—0.37mg/g,and 0.19—0.41mg/g,respectively,and those in spore powder were 0—0.01 mg/g,0,and 0—0.03mg/g,respectively.Conclusion The method is proved to be convenient and reliable,so it could be used for the quantitative analysis of ganoderic acids in Ganoderma.
