缺氧对大鼠骨髓间充质干细胞骨向诱导分化过程中骨钙素基因表达的影响

The effection of hypoxia on oteocalcin gene expression in rat bone marrow mesenchymal stem cells in course of differentiating into osteoblast

摘要 目的探索缺氧刺激对大鼠骨髓间充质干细胞(rat bone marrow mesenchyme stem cells,rBMSCs)骨向诱导分化过程中骨钙素(oteocalcin,OC)基因表达的影响。方法本研究把rBMSCs骨向诱导后按不同氧浓度及不同作用时间分组,检测不同作用时间的缺氧对rBMSCs骨向诱导分化过程中OC基因表达的影响。结果缺氧培养对rBMSCs骨向诱导分化中OC基因表达的影响在各时间段均有明显抑制作用且有统计学差别(P<0.05)。结论缺氧培养对rBM-SCs骨向诱导分化过程中OC基因表达抑制作用明显,表明2%缺氧对rBMSCs骨向诱导分化有抑制作用,促使骨改建平衡向破骨方向倾斜。 Objective To explore the effection of hypoxia on oteocalcin(OC) gene expression in rat bone marrow mesenchyme stemcells in course of differentiating into osteoblast.Method The hypoxia model of hBMSCs was devided into groups according to different oxygen density and different hours,and detect effects of different time on OC gene expression in the course.Results Hypoxia cultivanting can obviously inhibited OC gene expression of rat bone marrow mesenchymal stem cells in course of differentiating into osteoblast,and had statistically significant difference(P0.05).Conclusions Hypoxia cultivanting can obviously inhibit OC gene expression of rat bone marrow mesenchymal stem cells to differentiate into osteoblast.2% of hypoxia stimulus had obviously depressant effect to OC gene expression compared with the control group,which prompted bone remodeling equilibrium inclined bone fracture.
