摘要
目的观察聚焦超声致早孕小鼠子宫平滑肌及胚胎的急性损伤和转归效应。方法将96只妊娠第7天的小鼠随机分为A、B、C三组,每组32只,分别采用声强为0W/cm2、19W/cm2、32W/cm2的超声辐照小鼠妊娠子宫右侧,经超声辐照后即刻和继续妊娠第14天分别选取16只小鼠,取子宫平滑肌及胚胎,进行光镜、电镜观察及甲胎蛋白酶活性测定。结果经超声辐照后即刻光镜、电镜下显示B、C组胚胎细胞溶解,子宫平滑肌见可逆性损伤;继续妊娠第14天,B、C组胚胎细胞变性坏死,B组子宫增生较好,C组可见肌细胞溶解等改变。超声辐照后即刻B、C组子宫平滑肌Na+-K+-ATP、Mg++-ATP、Ca++-ATP酶活性均有升高趋势,与A组比较差异有统计学意义(P<0.05)。结论一定剂量的超声辐照早孕小鼠子宫,可致胚胎细胞变性坏死,而子宫平滑肌呈可逆性损伤。
Objective To observe the acute injury and outcome effect of focused ultrasound on uterine smooth muscle and embryos in mouse with early pregnancy.Methods Ninety-six mice at day 7 during the gestation were randomly divided into group A, B and C (n=32) and ultrasound exposure at the intensity of 0 W/cm2,19 W/cm2 and 32 W/cm2 were respectively performed on the fight side of uterus in pregnant mice.Sixteen mice were sacrificed immediately and uterine smooth muscle and embryos were dissected for histological observation and ATPase activity test.The rest 16 mice in each group were performed the same procedure 14 d later.Results After ultrasound exposure, cytolysis in embryos and reversibility injury in uterine smooth muscle were found in group B and C.On the 14th day after ultrasound exposure,degeneration and necrosis of embryonic ceils were found in both group B and C. Endometrium hyperplasia was found in group B,but eytolysis in smooth muscle was found in group C.After ultrasound exposure immediately, the Na+-K+-ATPase, Mg++-ATPase and Ca++-ATPase enzymatic activities had a trend of increasing in group B and C. Compared with group A, there was a significant difference(P 〈 0.05 ).Conclusion The ultrasound irradiation at certain intensity can cause degeneration and necrosis of embryos cells and reversible damage in uterine smooth muscle.
