摘要
采用牛津杯打孔法,以大肠杆菌、金黄色葡萄球菌、枯草芽孢杆菌、单核细胞增生李斯特菌、副溶血弧菌、米曲霉为指示菌,对从南海海域中捕捞的华贵栉孔扇贝肠道中分离到的9株乳酸菌进行抗菌活性测定,并对活性菌株的发酵液进行排酸、排过氧化氢,以及胰蛋白酶、木瓜蛋白酶、胃蛋白酶、蛋白酶K酶解实验,筛选出1株具有较强抑菌作用产细菌素的乳酸菌ZH-54。结合形态鉴定、生理生化特性实验以及16S rRNA序列同源性分析,鉴定菌株ZH-54为坚强肠球菌(Enterococcus durans),其16S rRNA在GeneBank的登录号为JX258806。
In this research, nine lactic acid bacteria strains were isolated from intestinal tract of a Chlamys nobilis from South China Sea. Their antibacterial activity to Escherichia coli, Staphylococcus aureus, bacillus subtilis, Listeria monocytogenes, vibrio prholyticus and Aspergillus oryzac were detected by Oxford plate double- layer agar experiment, and the antibacterial active strains were further studied to exclude the inhibitive effect of organic acid, H2Oz, and to observe the sensitivity of four proteases'. The research found that lactic acid bacterium ZH-54 was a bacteriocin-producing strain. ZH-54 was identified as Enterococcus durarts on the basis of morphological and physiological-biochemical characteristics and phylogenetic tree, and the accession number of 16s rRNA in GenBank was JX258806.
