摘要
目的 :为获得人血小板生成素全长cDNA克隆。方法 :用RT PCR技术 ,以特异性寡核苷酸为上、下游引物自胎肝总RNA中扩增目的基因。结果 :PCR扩增产物克隆至pUC18 T载体 ,获得重组质粒pUCT TPOM。 结论 :序列分析显示 。
Objective:To clone human thrombopoietin(TPO)cDNA.Method:RTPCR was used to amplify the target gene with total RNA from human fetal liver as template and specific oligonucleitides as upstream and downstream primers of TPO.Results:PCRamplified DNA fragment was successfully cloned into pUC18Tvector.Conclusion:DNA sequencing of recombinant plasmid containing human TPO cDNA indicated that cloned TPO cDNA sequence is the same with that reported previously.
