摘要
目的 :探讨p75NGFR介导神经细胞凋亡的分子机制。方法 :以转染了p75NGFR的大鼠小脑神经细胞株R2 L1 为细胞模型 ,应用酵母双向杂交系统 (two -hybridsystem)筛选和鉴定 p75NGFR胞内区p75ICD作用的蛋白。 结果 :和 p75NGFR作用的蛋白由一个 60 0bp大小的cDNA片段编码。结论 :该工作将有助于探讨p75NGFR介导细胞凋亡的分子机制和信号通路 ,为探索神经系统退行性病变的病因。
Objective:To explore the mechanism of p75 NGFR signals in neural cell apoptoic process.Methods:Two-hybrid system was used to screen a library for genes encoding a novel protein that interacted with the intracellular domain of p75 NGFR .The library was constructed by using cDNA from p75 NGFR -expressing R 2L 1 cells in a serum-free medium to induce apoptosis.Results:A 600 bp cDNA fragment encoded a novel protein that interacted with p75 ICD.Conclusions:The study is beneficial to exploring how the neurodegenerative disorders and brain aging occur,and how to control them.
