摘要
目的研究恩度联合化疗对直结肠癌细胞的抑制作用,并进一步观察不同Stat6表型的直结肠癌细胞对恩度联合化疗药物敏感性的差异。方法采用MTT法检测不同浓度的恩度单药或联合5-Fu及奥沙利铂对2种不同Stat6活化表型的直结肠癌细胞(Caco-2和HT-29)的增殖抑制作用。用流式细胞仪检测不同药物处理组对2种直结肠癌细胞早期凋亡率的影响。结果恩度单药作用于Caco-2和HT-29细胞没有显示出明显的增殖抑制和凋亡促进的作用(P>0.05)。而恩度联合1种或2种化疗药物[(5-Fu)和奥沙利铂(OXP)]对这2种癌细胞的生长抑制和凋亡促进的能力明显升高(P<0.05和P<0.01)。恩度联合化疗药物对Caco-2细胞生长抑制率和早期凋亡率均高于HT-29细胞(P<0.05)。结论恩度联合化疗药物能更好的抑制直结肠癌细胞的增殖能力,不同Stat6活化状态可能成为评价化疗疗效的潜在指标之一。
Objective To investigate the combined inhibitory effects of Endostar and chemotherapeutic drugs on growth and apoptosis of human colorectal cancer cell lines,and further observe the different chemosensitivity of these two cancer cells treated with Endostar combined with chemotherapeutic drugs in vitro.Methods The inhibitory rate of these two cancer cells was tested against the different concentration of Endostar(50,100,200,400 ug/ml respectively) combined with anti-colorectal cancer drugs 5-fluorouracil(5-FU,100 ug/ml) and oxaliplatin(OXP,45 ug/ml) using MTT assay.Their early apoptotic rates were examined by flow cytometry.Results The inhibitory rates and the early apoptotic rates didn't increased with the different concentration of Endostar both in human colorectal cancer cell lines HT-29 and Caco-2(P0.05).Endostar combined with one or two chemotherapeutic drugs(5-Fu and OXP) could obviously suppress the growth of HT-29 and Caco-2 and induce their apoptosis(P0.05 and P0.01).Compare to HT-29 cells,Caco-2 cells exhibited higher inhibitory rate and higher early apoptotic rates when Endostar combined with one or two chemotherapeutic drugs(P0.05).Conclusion The synergistic inhibitory effect on human colorectal cancer cells was strengthened by Endostar combined with chemotherapeutic drugs.Different activation phenotypes of Stat6 could become a potential marker to evaluate the biological characteristics of cancer cells and the curative effect of chemotherapeutic drugs and molecular targeting drugs.
