摘要 目的研究慢性丙型肝炎患者外周血纤维蛋白mRNA水平与肝纤维化的相关性。方法收集48例慢性丙型肝炎患者,全部进行肝穿刺活检,肝穿刺标本采用Masson染色及常规HE染色,判断肝脏炎症程度G和纤维化分期(S);分离患者外周血单个核细胞,抽提RNA,Real-time PCR检测FBRS mRNA表达水平,分析FBRS mRNA表达水平与肝脏炎症程度和纤维化分期的相关性。结果随着炎症活动度和纤维化程度的加重,外周血单个核细胞FBRS mRNA的表达水平逐渐升高,G分级和S分期均与外周血单个核细胞FBRS mRNA表达水平呈明显正相关r分别为0.593和0.826,P<0.01。结论外周血单个核细胞FBRS mRNA水平与肝脏炎症程度和肝纤维化程度密切相关,有可能成为评估HCV相关性肝纤维化程度的有效血清学指标。 Objective To investigate relationship between fibrosin mRNA expression in peripheral blood and hepatic fibrosis in chronic hepatitis C patients. Methods Forty-eight chronic hepatitis C patients were collected and treated by liver biopsy. The biopsy specimens were stained with HE and Masson stain. The hepatic inflammation grade (G) and hepatic fibrosis stage (S) were analyzed according to the standard of prevention and treatment scheme of viral hepatitis. FBRS mRNA expression level in peripheral blood mononuclear cells was examined by real-time PCR. The relationship among hepatic inflammation grade, hepatic fibrosis stage and FBRS mRNA expression level were analyzed. Results FBRS mRNA expression level in peripheral blood mononuclear cells was gradually in- creased with the development of hepatic inflammation grade and hepatic fibrosis stage. FBRS mRNA level in peripher- al blood was positively correlated with G and S (r=0.593 and 0.826, respectively, P〈0.01). Conclusion FBRS mRNA level in peripheral blood is closely related to the degree of hepatic inflammation and hepatic fibrosis, which might be one of useful indexes to evaluate the degree of liver fibrosis in chronic hepatitis C patients.
