5-HT受体亚型mRNAs在大鼠三叉神经节和脊髓背根节的表达——PCR法研究 被引量:13

摘要 根据 5- HT受体 13种亚型互补 DNA序列合成相应的特异性引物 ,用 PCR方法对各种 5- HT受体亚型在大鼠三叉神经节及脊髓背根节的表达进行了观察。结果显示 :三叉神经节表达 5- HT1A、5- HT1B、5- HT1D、5- HT2 A、5- HT3、5- HT4 和 5- HT7亚型。背根节表达的 5- HT受体亚型与三叉神经节者相同 ,但这些亚型的电泳条带密度在两种神经节存在着差异。三叉节的 5- HT1D和 5- HT3电泳条带密度最高 ,5- HT1A和 5- HT4 次之 ,5- HT1B、5- HT2 A和 5- HT7较弱 ;背根节的 5- HT3和 5- HT1D电泳条带密度最强 ,5- HT1B和 5- HT2 A次之 ,而 5-HT1A、5- HT4 和 5- HT7则较弱。将各亚型的 PCR阳性产物进一步克隆至 p CR 载体进行序列测定 ,结果表明这些 PCR产物的 DNA序列与已知的各亚型的序列一致。本研究的结果对进一步探讨 5- The method of polymerase chain reaction (PCR) was used to detect the presence of 5 HT receptor subtypes in the trigeminal ganglion (TG) and spinal dorsal root ganglion (DRG) of the rat. The specific oligonucleotide primers were synthesized based on the complementary DNA sequence for each of the 13 cloned rat 5 HT receptor subtypes. Expression of 5 HT 1A , 5 HT 1B , 5 HT 1D , 5 HT 2A , 5 HT 3, 5 HT 4 and 5 HT 7 receptor subtype mRNAs were detected winthin TG. DRG displayed banding identical to the TG. Positive band intensities for the various 5 HT receptor subtypes were different in TG and DRG. In TG, prominent bands were present for the 5 HT 1D and 5 HT 3 receptor subtypes and slightly less prominent bands were present for the 5 HT 1A , 5 HT 4, 5 HT 1B , 5 HT 2A and 5 HT 7 receptor subtypes. In DRG, prominent bands were present for the 5 HT 3 and 5 HT 1D receptor subtypes and slightly less prominent bands were present for the 5 HT 1B , 5 HT 2A , 5 HT 1A , 5 HT 4 and 5 HT 7 receptors subtypes. PCR products from each positively detected receptor subtype were subcloned into pCRⅡ vector for sequencing and found in correspond to published complementary DNA sequences. The present results might be significant for further analysing the role of 5 HT receptors in the primary sensory information transmission.
