昆虫细胞表达重组人胰岛素样生长因子1的研究 被引量:4

摘要 目的 利用杆状病毒 Ac MNPV载体在昆虫细胞中表达重组人胰岛素样生长因子 1( rh IGF- 1)。方法 含有胰岛素样生长因子 1( IGF- 1) c DNA的重组 Ac MNPV感染昆虫细胞株 Sf9及 Sf2 1后 ,采用酶联免疫吸附试验 ( EL ISA)检测被重组病毒感染的昆虫细胞表达产生的 rh IGF- 1,以野生型 Ac MNPV感染的昆虫细胞及空白细胞的细胞培养液、细胞裂解物作对照。结果  1重组 Ac MNPV感染的昆虫细胞大量表达有免疫活性的 rh IGF- 1,并可分泌至细胞培养液中 ;2受感染昆虫细胞 Sf2 1的培养上清中 rh IGF- 1含量较 Sf9高 ,而细胞裂解物则相反。结论 利用杆状病毒表达系统在昆虫细胞 Sf9和 Sf2 1中可大量表达 rh IGF- Objective To study expression of recombinant human insulinlike growth factor 1 (rhIGF1) in insect cells using baculovirus (AcMNPV) vector. Methods Insect cell lines of Sf9 and Sf21 were used as host cells for infection of recombinant IGF1 AcMNPV. The expressed rhIGF1 of the recombinant AcMNPV infected cells was measured quantatively with enzyme linkedimmune sorbent assay (ELISA). The media and cell lysates of wild type virus infected cell culture or uninfected cell culture were used as controls. Results ①rhIGF1 was highly expressed in both Sf9 and Sf21 cells after infected with recombinant virus, and secrected into culture media. ②rhIGF1 in Sf21 culture media was higher than that in Sf9 culture media. However, rhIGF1 in Sf21 cell lysate was lower than that in Sf9 cell lysate. Conclusion rhIGF1 can be successfully expressed in insect cells using AcMNPV baculovirus vector.
