摘要
为了建立检测非洲猪瘟病毒(ASFV)间接酶联免疫吸附试验(iELISA),利用杆状病毒表达载体pFastBac HTA在昆虫细胞Sf9中表达非洲猪瘟病毒的VP73蛋白,SDS-PAGE和Western blot检测显示重组蛋白大小约65ku,能与ASFV标准阳性血清发生特异性结合。以此重组蛋白作为检测抗原包被酶标板,初步建立了检测ASFV血清抗体的iELISA方法。表达的重组蛋白具有良好的特异性,为制备ASFV免疫血清学诊断检测试剂和疫苗提供了材料。
With the purpose of establishing an indirect ELISA (iELISA) method of African swine fever (ASF), recombinant protein VP73 was expressed in Sf9 insect cells with the baculovirus expression vector pFastBac HTA. SDS-PAGE and Western blot showed that the recombinant protein was about 65 ku and highly special to ASFV standard serum antibody. Coated with the recombinant protein, an iELISA method of ASF was established, which indicated the expressed protein was of good speeifici and would be useful material for studying diagnostic techniques and developing vaccine of ASFV.
