根霉K-1高产糖化酶固体发酵条件优化 被引量:2

摘要 以产糖化酶的根霉菌K-1为研究出发菌株,探讨根霉固体发酵生产糖化酶的最佳产酶条件。结果表明,此菌株产糖化酶的最佳培养基成分为:以麸皮为基本培养基,添加0.5%蛋白胨、干基重的1%尿素、0.05%KH2PO4、0.01%MgSO4.7H2O、0.05%CaCl2.2H2O,55%的含水量;最佳培养条件为接种量20%(干基计),培养温度为30℃,培养时间为36~48 h,该条件下产糖化酶的酶活力较高,最高酶活为549 IU/g干曲。 Rhizop us K-1 was used to produce glucoamylase through solid fermentation. The best culture mediums was composed of bran (as the basic carrier), 0.5 % peptone, urea 1% (dry base), 0.05 % KH2PO4, 0.01% MgSO4·7H2O, and 0.05 % CaCl2·2H2O. The best culture conditions were summed up as follows: inoculating quantity was 20 % (counted by dry base), culture temperature was at 30 ℃, and culture time was 36-48h. Under the above conditions, the produced glucoamylase had comparatively high activity as 549 IU / g(dry base).
