改良型Ogu-NWSUAF CMS油菜恢复性的遗传研究

摘要 为了揭示改良型Ogu-NWSUAF CMS的不育及恢复性的机理,以Ogu CMS油菜和Ogu-NWSUAFCMS油菜为母本,R1、中双2号为父本进行杂交,Ogu CMS与R1、中双2号的杂交F1均表现为不育,Ogu-NWSUAF CMS与中双2号的杂交F1表现不育,而与R1的杂交F1全可育,表明Ogu-NWSUAF CMS和Ogu CMS的恢保关系不同。利用一步四重PCR方法对Ogu-NWSUAF CMS和Ogu CMS进行扩增,发现Ogu-NWSUAF CMS既能扩增出Ogu CMS orf 138基因的特征带,又能扩增出Nap CMS orf 222基因的特征带,表明Ogu-NWSUAF CMS与Ogu CMS分子特征不同。以恢复材料R1与Ogu-NWSUAF CMS杂交、自交、测交获得相应的F1、F2和BC1,对F1、F2、BC1的育性进行调查,并使用χ2c测验相应的分离比例,在BC1群体中可育株与不育株的分离比符合1∶1(χ2c=0.175<χ20.05(1)=3.84),在F2群体中,虽然χ2c测验可育株与不育株的分离比在理论上不符合3∶1与15∶1两种分离比,但更为接近3∶1,远远偏离15∶1,故Ogu-NW-SUAF CMS恢复性的遗传符合一对显性核基因控制的模式。 In order to reveal the sterile and restoration mechanism of improved Ogu-NWSUAF CMS,in this study Ogu CMS and Ogu-NWSUAF CMS rapeseed were used as the female to testcrossed with R1 and Zhongshuang 2,respectively.The testcross offsprings of Ogu CMS with R1,Zhongshuang 2 showed sterility.The testcross offsprings of Ogu-NWSUAF CMS with Zhongshuang 2 was sterility,while testcross offsprings with R1 was fertility.This suggested that the restoring and maintaining relationship between the Ogu-NWSUAF CMS and Ogu CMS was different.A multiplex PCR assay showed that the characteristic fragments both of Ogu CMS system specific gene-orf138 product and Nap CMS system specific gene-orf 222 product could be amplified from Ogu-NWSUAF CMS,which indicated that the molecular characteristics of Ogu-NWSUAF CMS was different from that of Ogu CMS.The restorer R1 was crossed with the Ogu-NWSUAF CMS and resulted in F1,F2 and BC1 populations.According to the Chi-square test,fertility segregation ratio in BC1 populations was agree with 1∶1(fertility∶sterility,χ2c=0.175χ20.05(1)=3.84).While fertility segregation ratio in F2 population was not agree with 3∶1 or 15∶1(fertility∶sterility),farther from 15∶1,but was closer to 3∶1.Therefore,The restoration of Ogu-NWSUAF CMS was controlled by a pair of dominant nuclear gene.
