药物诱导的小鼠红白血病细胞沿红系分化过程中基因表达的差异显示分析

摘要 应用mRNA差异显示技术分析了药物诱导MEL细胞沿红系分化过程中基因表达的改变。以六个锚定引物和六个随机引物共36种组合进行差异显示分析,得到67个差异片段。对部分片段进行克隆测序,获20个片段的序列,其中12个新的表达序列标签。以反向斑点杂交检测部分差异显示带的差异性,阳性率为27/39(69%)。 The mRNA differential display was adopted to analyze the differences of gene expression in murineerythroleukemia cells before and after chemical-induced eothroid differentiation. After differential display analysisunder 36 combinations with six anchored primers and 6 random primers, 67 interest bands were selected, and 21cDNA fragments were cloned into T-vector and sequenced, with 12 new ESTs identified. The reverse Northernblotting analysis was used to further characterize the expression difference of 21 selected interest bands with 60%positive ratio finally.
