摘要
应用mRNA差异显示技术分析了药物诱导MEL细胞沿红系分化过程中基因表达的改变。以六个锚定引物和六个随机引物共36种组合进行差异显示分析,得到67个差异片段。对部分片段进行克隆测序,获20个片段的序列,其中12个新的表达序列标签。以反向斑点杂交检测部分差异显示带的差异性,阳性率为27/39(69%)。
The mRNA differential display was adopted to analyze the differences of gene expression in murineerythroleukemia cells before and after chemical-induced eothroid differentiation. After differential display analysisunder 36 combinations with six anchored primers and 6 random primers, 67 interest bands were selected, and 21cDNA fragments were cloned into T-vector and sequenced, with 12 new ESTs identified. The reverse Northernblotting analysis was used to further characterize the expression difference of 21 selected interest bands with 60%positive ratio finally.
