摘要
目的探讨蛋白酶体抑制剂硼替佐米(Bortezomib,商品名:万珂)对K562细胞株DNA甲基转移酶(DNA methyltransferase,DNMTs)表达、细胞凋亡的影响。方法常规体外培养白血病K562细胞株,随机将处于对数生长期的细胞分为12h、24h、36h 3个作用时间组,予以不同浓度的硼替佐米:0,6,20,60nmol·L-1,Western印迹法检测胞内DNMT1的表达,流式细胞术检测细胞凋亡。结果与阴性对照组相比,硼替佐米可显著抑制DNMT1表达。硼替佐米作用于细胞12、24、36h后细胞凋亡率逐渐增加,60nmol·L-1硼替佐米作用36h后细胞凋亡率为(61.68±3.20)%;结论硼替佐米抑制K562细胞株DNMT1表达,诱导细胞凋亡,该作用呈浓度依赖性。
OBJECTIVE To study the activity of DNA methyltransferase(DNMTs) in K562 cell line exposed in Bortezomib,and the role of Dnmts in the process.METHODS K562 cells were treated with bortezomib of the concentrations of 0、6、20 and 60nmol·L-1 respectively,Western-blot was used to analyse the intracellular expression of DNMT1.Flow cytometry was used to detect apoptosis.RESULTS Which was(61.68±3.20)% after treating with nmol·L-1 Bortezomib could induce the apoptosis of K562 cells.After treating with Bortezomib for 12 hours、24 hours、36 hours respectly,the apoptosis rate increased gradually.CONCLUSION Bortezomib can inhibits DNMTI expression and induces k562 cellsapoptosis,which shaws concentration-dependent manner.
