摘要
目的观察益气血补肝肾方对整合素ανβ3在早孕小鼠子宫内膜中表达的影响。方法采用免疫组织化学定位观察整合素ανβ3在早孕小鼠子宫内膜中的表达;采用实时荧光定量PCR在mRNA水平上检查整合素ανβ3在早孕小鼠子宫内膜中的表达。结果免疫组化结果显示,整合素ανβ3蛋白在妊娠早期小鼠表达于子宫内膜腔上皮、腺上皮细胞的细胞膜及胞浆中,呈棕黄色颗粒状。D1、D2表达最弱,D4表达明显增加,D5表达最强,D6表达减弱;实时荧光定量PCR结果显示,整合素ανβ3在中药组和对照组早孕小鼠子宫内膜中的表达随妊娠天数呈规律变化,在中药组的表达高于对照组,但差异无统计学意义。两组中D4和D5表达最强,与D1、D2、D6比较差异具有显著性(P<0.05),整合素αν在D5的表达显著高于D4(P<0.01)。结论益气血补肝肾方可改善子宫内膜的容受性,促进胚胎与子宫内膜发育的同步性,从而在胚胎着床过程中发挥重要作用。
Objective To observe the effect of Yiqixuebuganshen decoction on integrin ανβ3 in endometrium of the early pregnant mice.Methods Immunohistochemistry and realtime RT-PCR were used to detect the expression of integrin ανβ3 in endometrium of the early pregnant mouse.Results Immunohistochemistry showed that integrin ανβ3 expressed in the mambrane and cytoplasm of endometrial luminal epithelium and epithelial of the early pregnant mice.It weakly expressed on D1 and D2,significantly expressed on D4,achieved to the peak on D5;Realtime RT-PCR showed that integrin ανβ3 expressed regularly in the endometrium of medicine and control groups.It expressed stronger in medicine groups than in control groups,but the differences are no significance（P0.05）.In the two groups,it was significantly higher expressed on D4 and D5 than that on D1、D2 and D6（P0.05）.Integrin αν expressed more strongly on D5 than on D4（P0.01）.Conclusion Yiqixuebuganshen recipe is very important to improve endometrial receptivity and play an important role in embryo implantation.
