针刺对脑卒中痉挛大鼠模型大脑皮质DRD1mRNA、DRD2mRNA表达的影响 被引量:19

摘要 目的通过观察针刺对脑卒中痉挛状态大鼠大脑皮质多巴胺受体亚型(DRD1、DRD2)mRNA表达的影响,探讨针刺缓解脑卒中痉挛状态的作用机理。方法采用大脑中动脉线栓法制作大鼠脑卒中痉挛状态模型,常规手法针刺组(模型+常规手法针刺阳陵泉、曲池)及电针组(模型+电针阳陵泉、曲池)。以神经系统损伤症状、肌张力评分评价模型和针刺疗效,RT-PCR法观察大鼠大脑皮质DRD1mRNA、DRD2mRNA的表达。结果 (1)与模型组比较,针刺可降低大鼠神经系统损伤评分及痉挛大鼠的肌张力,比较差异有显著统计学意义(P<0.01),但常规手法针刺组与电针组比较差异无统计学意义(P>0.05);(2)大脑皮质DRD1mRNA、DRD2mRNA的表达:与模型组比较,常规手法针刺组与电针组表达均明显增加,比较差异有统计学意义(P<0.05,P<0.01);常规手法针刺组与电针组比较差异无统计学意义(P>0.05);结论针刺对脑卒中痉挛性瘫痪具有较好的疗效,其作用机理可能与调节中枢神经系统大脑皮质DRD1mRNA、DRD2mRNA的表达有关;但常规手法针刺与电针差异不明显。 Objective To explore the mechanism of acupuncture on relieving stroke spasticity by observing acupuncture treatment methods' influence on the Dopamine receptors expression of DRDlmRNA, DRD2mRNA cerebral cortex in spastieity model rats. Methods 40 Experimental SD rats were randomly divided into blank control group, sham operation group,model group,conventional handy acupuncture group (modeling + routine handy acupuncture on Yanglingquan and Quchi), electroacupuncture group (modeling + electroacupuncture on Yanglingquan and Quchi).Use the method of suture-occlusion to prepare stroke paralysis model in rats.Use the Nervous sysem damage symptom score and Spasticity assessment score to evaluate model and the effect of acupuncture, use the ELISA to analyse acupuncture treatment methods' influence on DA, DRD1, DRD2 contents in substantia nigra and cerebralcortex which of rats with cerebral stroke spastic paralysis, use the reverse transcription-PCR to observe acupuncture treatment methods' influence on DRDlmRNA, DRD2mRNA expression of cerebral cortex in spastieity model rats. Results (1) Acupuncture can decrease the Nervous system damage symptom score and the Muscle tension (P〈0.01); there was no significant difference between ceconventional handy acupuncture group and electroacupuncture group (P〉0.05). (2) DRDlmRNA, DRD2mRNA expression in cerebral cortex : Compared with model group, expression of conventional handy acupuncture group andcelectroacupuncture group were increased significantly, the difference was statistically significant (P〈0.05, P〈0.01); there was no significant difference between conventional handy acupuncture group and celectroacupuncture group (P〉0.05) . Conclusion Acupuncture has a good effect on the spasticity ofparalysed limbs after stroke, it' s function mechanism is may relative to ajusting the DA receptors expression in substantia cerebralcortex.
