蛋白酶激活受体-2与抑癌基因P53、人表皮生长因子受体-2在乳腺癌组织中的表达及其意义

摘要 目的探讨蛋白酶激活受体-2(PAR-2)、抑癌基因P53和人表皮生长因子受体-2(HER-2)在乳腺浸润性导管癌组织中的表达及其与临床病理组织学指标的关联。方法应用组织芯片和免疫组化法检测76例乳腺浸润性导管癌组织中蛋白的表达,采用χ2检验或Fisher精确概率法分析蛋白表达的关系。结果 76例癌组织中PAR-2阳性率为84.2%,P53为43.4%,HER-2为55.3%。PAR-2的表达与年龄、临床分期、肿瘤直径、组织学分级均无关,差异无统计学意义(P>0.05),与淋巴结转移有关,差异有统计学意义(P=0.022)。HER-2的表达与年龄、临床分期、肿瘤直径、淋巴结转移无关,差异无统计学意义(P>0.05),与组织学分级有关,差异有统计学意义(P=0.034)。P53的表达与年龄、肿瘤直径、组织学分级、淋巴结转移无关,差异无统计学意义(P>0.05),与临床分期有关,差异有统计学意义(P=0.03)。PAR-2与P53联合或与HER-2联合的阳性表达均与组织学分级有关,差异均有统计学意义(P<0.05)。PAR-2与HER-2表达有关,差异有统计学意义(P=0.001)。结论 PAR-2,P53,HER-2蛋白的异常表达可能涉及乳腺浸润性导管癌的发生和发展。 Objective To investigate the expression of protease activated receptor-2(PAR-2),P53 and human epidermal growth factor receptor-2(HER-2) in breast invasive ductal cancer tissues,and to explore the correlation between the expression of these proteins and clinical histopathological data.Methods The method of immunohistochemical staining was performed by tissue microarray within 76 specimens of invasive breast cancer.The chi-square test and Fisher test were used to analyze the correlation of the expression of the proteins.Results Positive expression rates of PAR-2,P53 and HER-2 in 76 cases were 84.2%,43.4% and 55.3% respectively.The expression of PAR-2 was non-related to age,clinical stage,tumor size and histopathological grade,the differences were not statistically significant(P 0.05),but it was correlated to the lymph node metastasis,the difference was statistically significant(P = 0.022).The expression of HER-2 was non-related to age,clinical stage,tumor size and lymph node metastasis,the differences were not statistically significant(P 0.05),but it was correlated to the histopathological grade,the difference was statistically significant(P = 0.034).The expression of P53 was non-related to age,clinical stage,tumor size and lymph node metastasis,the differences were not statistically significant(P 0.05),but it was correlated to the histopathological grade,the difference was statistically significant(P = 0.03).The combination of PAR-2 and P53 or HER-2 was associated with histopathological grade,the differences were statistically significant(P 0.05).The expression of PAR-2 was correlated with HER-2,the difference was statistically significant(P = 0.001).Conclusion PAR-2,P53,HER-2 may involve in the occurrence and development of breast invasive ductal cancer.
