探讨NDUFA13抑制结肠癌细胞增殖的机制 被引量:1

An investigation on the mechanism for NDUFA13 to inhibit proliferation of human colon cancer

摘要 目的探讨抑癌基因Ⅰ型辅酶脱氢酶1α亚复合物13(NDUFA13)抑制结肠癌生长的机制。方法以人结肠正常组织为对照,用实时荧光定量PCR及免疫组织化学方法检测NDUFA13及miR-7在60例人结肠癌中的表达;体外细胞模型研究NDUFA13基因对结肠癌细胞株增殖水平的影响。结果与人结肠正常组织比较,NDUFA13及miR-7在人结肠癌表达显著下调(P<0.01),两者的表达呈正相关;在人结肠癌细胞株SW480及HCT-116中高表达NDUFA13能够显著抑制结肠癌细胞的增殖。结论抑癌基因NDUFA13在人结肠癌中通过促进miR-7的成熟而抑制肿瘤的发生发展。 Objective To investigate the mechanism for NADH dehydrogenase 1 alpha subcomplex 13 (NDUFA13) to inhibit the proliferation of human colon cancer. Methods The expressions of NDUFA13 and miR-7 were detected by RQ-PCR and immunohistochemisty in 60 human colon cancers and normal colon tissues. The effect of NDUFA13 on the proliferation of colon cancer cells lines of SW480 and HCT-116 in vitro was tested. Results The expressions of NDUFA13 and miR-7 were both significantly downregulated in human colon cancer tissues compared to normal colon tissues (P^0. 01). The expression of NDUFA13A was positively correlated with that of miR-7. High-expressed NDUFA13 in SW40 and HCT-116 inhibited the proliferation of those. Conclusion NDUFA13 can inhibit the development and progression of human colon cancer by promoting mature of miR-7.
