摘要
目的:探讨放射性粒子125I联合黄连素对荷人肝癌裸鼠肝癌组织的病理变化影响。方法:建立人肝癌Hep-G2细胞移植瘤模型的Balb/c雌性裸鼠24只,随机分成4组,每组6只,A组给予生理盐水+空心125I粒子、B组给予黄连素45 mg/kg+空心125I粒子、C组给予生理盐水+125I粒子、D组给予黄连素45 mg/kg+125I粒子。观察各组小鼠肿瘤生长情况,应用光学显微镜和电子显微镜观察肝癌细胞的变化情况。结果:D组肿瘤体积最大抑制率为77.62%,与其他组比较差异统计学意义(P<0.01);电子显微镜下可见D组肿瘤细胞核染色质浓缩并边缘化,核碎裂,胞质空泡化明显。结论:黄连素能增强放射性125I粒子诱导肿瘤细胞凋亡能力,使肿瘤变性、坏死。
Objective: To investigate the pathological change of HEP-G2 Cells induced by radioactive particle 125 I combine with Berberine.Methods: Established model of liver cancer on24 Balb/c Female Nude Mice,and randomly divided into 4 groups(n=6): Group A,given to implantation of blank particles and daily intragastric administration of NS;group B,given implantation of a blank particles and 45mg·kg-1·d-1 orally berberine solution;group C,was given 125 I implantation of a particle(0.8 mCi) according to TPS and daily gived of;group D,was given 125I implantation of a particle(0.8 mCi) according to TPS and 45 mg·kg-1 orally berberine solution;The size of tumors was measured every 4 days and also the sensitivity coefficient were calculated.The ultrastructural changes of HEP-G2 cell after 125 I particles combined with Berberine treatment were examined with transmission electron microscope(TEM).Results: The maximum tumor inhibition rate of treated group was group D,and the inhibition rate was 77.62%,there was significant difference from the other groups(P0.01).Condensation of chromatin at margins of nuclei,disintegration of nucleolus,vacuoles in cytoplasm were observed in group D under TEM.Conclusion: The interstitial implant of radioactive seed 125I induces hepatoma cells apoptosis. 125I combined with Berberine induces apoptosis,cell degeneration and necrosis.
