摘要
目的探讨夏枯草联合山慈姑对甲状腺癌细胞的抗增殖作用。方法不同浓度的夏枯草和山慈姑作用于人甲状腺癌SW579细胞48 h后,采用MTT法分别计算出夏枯草和山慈姑对SW579的半数抑制浓度(IC50);实验分为夏枯草组、山慈姑组、夏枯草与山慈姑联合处理组及阴性对照组,将各组药物分别作用于SW579细胞48 h后,倒置相差显微镜下观察各组药物作用于SW579后细胞的形态学变化;Western blot检测各组c-myc蛋白的表达。结果夏枯草对SW579细胞的半数抑制浓度IC50为21 mg/mL,山慈姑的半数抑制浓度IC50为122 mg/mL;夏枯草组与联合组能抑制SW579细胞的生长,且联合组的抑制作用较夏枯草组明显;c-myc蛋白在夏枯草组及联合组的表达均较阴性对照组低,且在联合组的表达较夏枯草组表达更低(P<0.05),小剂量山慈姑组与阴性对照组比较差异无显著性(P>0.05)。结论夏枯草联合小剂量山慈姑对甲状腺癌细胞增殖的抑制作用较为明显,为中药联合治疗甲状腺癌提供理论依据。
【Objective】 To investigate the anti-proliferation effect of traditional Chinese medicine prunella vulgaris and cremastra appendiculata on the thyroid cancer cell line in vitro.【Methods】 The human thyroid cancer cell line SW579 was incubated particularly with different concentrations of Prunella vulgaris and Cremastra appendiculata,then the inhibition concentration 50%(IC50) of each drug can be measured by MTT assay.It can be divided into four groups: the Prunella aulgaris group,the Cremastra appendiculata group,both of the two drugs and negative group.The change of the cell morphology can be observed by inverted phase contrast microscope after treating with different drugs.The expression of c-myc protein in different drug therapeutic teams was detected by Western blot.【Results】 The IC50 value of Prunella vulgaris was 21 mg/mL,and the IC50 value of Cremastra appendiculata was 122 mg/mL.The Prunella vulgaris and the two drugs group can inhibit the cell growth,especially in the two drugs group.The c-myc protein of the Prunella vulgaris group and the two drugs group were lower than the negative group(P 0.05),especially in the two drugs group(P 0.05),there is no difference between the small dose of Cremastra appendiculata and the negative group(P 0.05).【Conclusions】 The traditional Chinese medicine Prunella vulgaris and cremastra appendiculata can anti-proliferate the growth of SW579 cell line,the small dose of cremastra appendiculata can promote the anti-proliferation effect of prunella vulgaris.
