摘要
采用肾上腺素法测定了超氧化物歧化酶的酶活,确定了最佳测定波长,并研究各种实验条件对超氧化物歧化酶活性的影响。结果表明最优的测定酶活条件为:测定波长为490 nm,实验温度为30℃,0.05 mol·L-1碳酸钠-碳酸氢钠内含1×10-4mol·L-1乙二胺四乙酸缓冲溶液(pH为10.2),肾上腺素浓度为3×10-4mol·L-1,控制酶的加入量为0.30 mL。
SOD activity was measured by epinephrine autoxidation method. The optimum wavelength was obtained. In addition, the effects of different experimental conditions on SOD activity were studied. The results indicated that the optimum conditions were as follow, constituents and concentration of the buffer: the optimum wavelength 490 nm, the temperature 30 ℃, 0.05 mol.L^-1 Na2CO3-NaHCO3 buffer containing 1 ×10^-4mol·L^-1IEDTA (pH=10.2), the concentration of epinephrine: 3×10^-4mol·L^-1; control enzymes adding level at 0.3 mL.
