摘要
目的:通过观察糖肾康饮对实验性糖尿病肾病大鼠肾组织糖基化终产物(AGES)、转移生长因子"1(TGF-"1)的表达沉积,肾功能及肾脏结构的影响,进一步明确糖肾康饮治疗糖尿病肾病的作用机制。方法:切除大鼠左侧肾脏联合腹腔注射链脲佐菌素(50mg/kg)建立糖尿病肾病(DN)模型,90只雄性Wistar大鼠随机分为空白组、模型组、西药马来酸依那普利组、中药糖肾康饮高,中,低剂量组,给药12周后测血尿指标,取右侧肾脏,采用免疫组化法检测肾组织AGES和TGF-"1的表达。结果:模型组大鼠肾组织AGES和TGF-"1大量表达,镜下特征表现为肾小球细胞外基质(ECM)积聚所致肾小球基底膜、系膜增生,经12周的糖肾康饮治疗后肾组织中AGES和TGF-"1明显少于模型组,以糖肾康饮高剂量组效果最好,但与中剂量组相比无显著差异。结论:糖肾康饮能够减少ECM积聚,降低DN大鼠肾组织AGES和TGF-"1的表达,可能是通过降低AGES和TGF-"1的表达,减少肾小球细胞外基质(ECM)积聚而改善DN肾脏病理结构。
Objective: To investigate the effect of Tangshenkang produced on DN by observing the expression of AGES and TGF-β1of renal tissue in STZ-induced Diabetic Nephropathy Rats.Methods: The rat models were induced by right nephrectomy and peritoneal injection of some dosage streptozotocin(50mg / kg).Ninety male Wistar rats were randomly divided into six groups: normal group,model group,Yina Puli group,Tangshenkang high-dose,middle-dose,low-dose treatment group,and each group with ten rats.After 12 weeks of treatment,test the blood and urinary guidelines and finish the nephrectomy.The expressions of AGES and TGF-β1 in the kidney were detected by immunohistochemistry.Results: The AGES and TGF-β1 expressed highly in the kidneys of the model rats,ECM accumulated when it was observed with microscope,while lower in the kidneys which were treated by Tangshenkang,and expressed lowest in the high-dose group,but there was no significant difference from middle-dose group.Conclusion: The Tangshenkang formula can decrease the accumulation of ECM and the expression of AGES and TGF-β1 in the kidney of DN rats,and it may be the mechanism of Tangshenkang on reducing ECM accumulation and improving the DN kidney pathology.
