摘要
目的:探讨血清高迁移率族蛋白1(HMGB1)水平与心衰的相关性;评价辛伐他汀对心衰大鼠心功能不全的保护效应。方法:采用肾上腹主动脉缩窄法制作大鼠慢性心衰模型。60只大鼠随机分为心衰模型组及心衰辛伐他汀于预组和假手术组,每组各20只。用酶联免疫吸附法检测各组血清HMGB1、肿瘤坏死因子α(TNF-α)、白介素1(IL-1)水平,用多导生理记录仪记录血流动力学指标[中心静脉压(MAP)和左室舒张末压(LVEDP)]。处死动物后取心脏,称重左心室,计算左心室重量指数。结果:①与假手术组相比,心衰模型组大鼠HMGB1、TNF-α、IL-1明显升高(P<0.05);心衰辛伐他汀干预组和心衰模型组相比,HMGB1、TNF-α、IL-1均有明显改善(P<0.05)。②与假手术组相比,心衰模型组MAP有明显下降(P<0.05),而LVEDP明显升高(P<0.05);心衰辛伐他汀干预组和心衰模型组相比,MAP、LVEDP均有明显改善(P<0.05)。③与假手术组相比,心衰模型组左心室重量指数增加(P<0.05),心衰辛伐他汀干预和心衰模型相比,左心室重量指数下降(P<0.05)。结论:HMGB1可能参与了心力衰竭的过程,应用辛伐他汀可以降低心衰大鼠HMGB1水平并且改善心功能。
Objective: To explore the correlation of serum levels of high mobility group box chromosomal protein B1(HMGB1) with heart failure,to evaluate the protective action of simvastatin for heart function of rats with heart failure.Method: Rat models of chronic heart failure were established by constriction of abdominal aorta above kidney.Sixty rats were randomly divided into heart failure model group,heart failure with simvastatin intervention group(simvastatin intervention group) and false operation group,each with 20 rats.Serum levels of HMGB1,TNF-α and interleukin 1(IL-1) were examined by ELISA,and hemodynamic indices including systolic blood pressure(SBP),diastolic blood pressure(DBP),mean arterial pressure(MAP) and left ventricular end diastolic pressure(LVEDP) were examined with multi-lead physiological recorder.Heart of rat was taken and weighted after sacrifice,and weight index of left ventricle had been calculated.Result: ①In comparison with false operation group,serum levels of HMGB1,TNF-α and IL-1 were significantly elevated in rats of heart failure group.②In comparison with false operation group,MAP was significantly decreased(P0.05) and LVEDP was significantly elevated(P0.05) in rats of heart failure group.MAP and LVEDP were significantly improved in rats of simvastatin intervention group compared with those of heart failure group(P0.05).③In comparison with that of false operation group,left ventricular weight index in rats of heart failure group was increased(P0.05),and left ventricular weight index was decreased in rats of simvastatin intervention group compared with that of rats in heart failure group(P0.05).Conclusion: It is probable that HMGB1 may be participated in the process of development of heart failure,the application of simvastatin can decrease the level of HMGB1 and improve the function of heart.
