实时多聚酶链式反应验证人类原发性膝关节骨性关节炎病变软骨基因表达的变化 被引量:2

摘要 目的:以关节软骨为研究对象,通过全基因组基因谱的表达,从上调3倍以上和下调3倍以上差异表达基因中任意选定10个基因进行复孔的PCR实时定量检测,验证所获得的差异基因在原发性KOA病理过程中的意义。方法:对12例原发性膝关节骨性关节炎病变软骨与4例正常膝关节软骨样本分别进行实时多聚酶链式反应。结论:通过3次重复试验,差异表达基因中任意选定的10个基因,经统计学处理,t检验,P<0.001。有显著性差异,证明膝关节软骨中的差异表达基因在原发性膝关节骨性关节炎发生过程中的关键作用。 Objective:In this study,the articular cartilage was as the research object, through the expressions of whole genome genetic spectrum, among up - regulating more than 3 times and down - regulating more than 3 times arbitrary dif- ferentially expressed genes, there were 10 genes selected to get real - time quantitative detection of PCR hole, and the differences in the genes obtained in the process of primary KOA were used to prove the pathological significance. Meth ods:12 cases of primary knee osteoarthritis cartilage and four normal knee cartilage samples,were given real -time poly merase chain reaction respectively. Conclusion:Through the three times repeated experiments, differences in the selected 10 genes had significant difference,P 〈0. 001. It is proved that the knee joint cartilage differentially expressed genes are the key in primary knee osteoarthritis process.
