摘要
为了快速建立芦笋全雄品种制种田,对全雄新品种WF-8的父母本组培快繁和过渡移栽技术进行了系统研究。结果表明:适合全雄品种WF-8母本启动成苗的培养基配比为6-BA1.0mg.L-1+NAA 0.1mg.L-1,适合父本启动成苗的培养基配比为6-BA1.0mg.L-1+NAA0.05mg.L-1;适宜父本增殖的培养基为MS+6-BA1.0mg.L-1+NAA 0.1mg.L-1,适宜母本增殖的培养基为MS+6-BA1.0mg.L-1+NAA 0.05mg.L-1+KT 0.1mg.L-1;蔗糖浓度由30g.L-1增加到35g.L-1,琼脂浓度由4g.L-1增加到4.5g.L-1,可以防止芦笋试管苗玻璃化;移栽成活率高达95.3%。
In order to quickly build asparagus male varieties for farm,the study on culture callus and transitional cultivating of a new all male asparagus hybrid "WF-8" parents was studied in detail. The results showed that: the suitable hormone proportion of female parent initiation culture callus was NAA 0. 1 mg. L-1 + 6-BA 1.0 mgoL l ,the suitable hormone proportion of male parent initiation culture callus was NAAO. 05 mg-L-1 + 6-BA1.0 mg·L^-1 The suitable proliferation culture medium for male parent was MS+6-BA1.0 mg·L^-1 + NAA 0.1 mg·L^-1 ,the suitable proliferation culture medium for female parent was MSq-6-BA 1.0 mg·L^-1 h-NAA 0.05 mg·L^-1 +KT 0.1 mg·L^-1;1 The sucrose concentration increased from 30 g·L^-1 to 35 g·L^-1 and the agar concentration increased from 4 g·L^-1 to 4.5 g·L^-1 could prevent the asparagus tissue culture seedling from vitri- ficatiom The survival rate of transolanting was 95.3%.
