摘要
目的探讨15-脱氧前列腺素J2(15 d-PGJ2)对非酒精性脂肪性肝病(NAFLD)大鼠的保护作用。方法将50只雄性Wistar大鼠随机分为正常对照组(NC组)和模型组,分别给予普通饲料和高脂饲料喂养,12周末制成NAFLD大鼠模型;再将模型组随机分为4组:15 d-PGJ2低剂量治疗组(LT组),15 d-PGJ2高剂量治疗组(HT组),模型对照组(MC组),饮食疗法组(DT组)。治疗2周后处死大鼠,观察各组大鼠的肝组织病理变化,检测血清丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST)、碱性磷酸酶(ALP)、空腹血糖(GLU)、甘油三酯(TG)、总胆固醇(TC)的水平。结果与MC组大鼠相比,LT组、HT组及DT组大鼠血清ALT、AST、ALP、GLU、TG、TC的水平降低,肝细胞脂肪变性及炎症浸润均有减轻,其中以LT组改变最明显(P<0.05)。结论适量的15d-PGJ2对NAFLD大鼠有明显的保护作用,可以改善大鼠的NAFLD。
Objective To study the protection of 15 - Deoxy -/X 12,14 -- prostaglandin J2 ( 15d - PGJ2) on nonalcoholic fatty liver disease (NAFLD) in rats. Methods Fifty male Wistar rats were randomly divided into normal control group ( NC group) and model group. The rats in NC group and model group were fed with ordinary feed and high - lipid feed respectively for 12 weeks to induce NAFLD in model group. The rats in model group were then further divided into low dosage 15d -PGJ2 treatment group (LT group) , high dosage 15d -PGJ2 treatment group( HT group), model control group( MC group), and ordinary diet treatment group ( DT group). Two weeks later, all rats were killed. Hepatic morpho- logical changes were observed under light microscope, the liver function parameters ( ALT, AST, ALP) , triglyceride (TG) , total cholesterol ( TC ) and glucose (GLU) of blood were examined by biochemistry methods. Results Compared with model group, hepatic steatosis and inflammatory in- filtration were palliated, the contents of ALT, AST, ALP, TC, TG, and GLU in serum were decreased in LT group, HT group and DT group. The above indexes were deereased most obviously in LT group ( P 〈 0.05 ). Conclusion The appropriate amount of 15d - PGJ2 has a good protective effect and improvements on NAFLD in rat.
