摘要
建立了快速测定苯巴比妥含量的方法。在pH 6.0的Clark-Lubs(C-L)缓冲溶液中,向一定浓度十六烷基三甲基溴化铵中加入荧光桃红,体系荧光增强,再加入苯巴比妥,体系荧光强度降低,且药品加入量与降低程度呈良好线性关系。在优化试验的条件下,线性范围是0.008~0.72 mg/L,检出限为0.02 mg/L。由该体系检测了药品在动物血清中的含量,回收率为93.6%~97.3%,相对标准偏差在2.3%~3.2%之间。建立了荧光桃红-十六烷基三甲基溴化铵测定苯巴比妥含量的荧光探针法。
To develop a new fast and simple method for the determination of phenobarbital(PB).Method: In the Clark-Lubs(C-L) buffer solution at pH 6.00,the fluorescence intensity of phloxine(PHLO) was enhanced when a certain concentration of cetyl trimethyl ammonium bromide(CTMAB) was added.The enhanced fluorescence intensity declined obviously after the addition of phenobarbital,and the declined fluorescence intensity was proportional to the concentration of phenobarbital.Under optimal experimental conditions,the linear range is 0.008-0.72 mg/L,and the detection limit is 0.02 mg/L.This method was applied to the detection of the content of phenobarbital in the serum of animal,and the average recoveries of the method ranged from 93.6% to 97.3% with the relative standard deviations(RSD) of 2.3%~3.2%.Phloxine-cetyl trimethyl ammonium bromide was used as the fluorescence probe in rapid determination of phenobarbital content,and this method is simple,accurate,rapid and highly sensitive.
