摘要
将椰壳活性炭进行可控氧化及硅烷偶联剂(APTMS)反应后,与戊二醛交联,成功地共价固载了木瓜蛋白酶.木瓜蛋白酶固载量为15 mg/g,固载酶的最佳活性pH值为7.4,温度为50℃.与游离酶相比,固定化酶的温度、pH稳定性明显提高.溶液酶和固定化酶的K_m分别为0.86 mol/L和0.95 mol/L,V_m分别为0.603和0.528 mol·s^(-1)·g^(-1).
Papain was successfully immobilized onto coconut shell activated carbons subjected to successive oxidation, aminosilylation and aldehyde grafting modification. The amount of immobilized papain on ACs was 15 mg/g. The immobilized enzyme showed maximum activity at a temperature of 50℃ and pH 7. 4. The immobilized enzyme has an increased resistance against the changes of temperature and pH in comparison to free enzyme. Km values were found to be 0. 86 mol/L and 0. 95 mol/L for the immobilized enzyme and free enzyme, respectively. The immobilized enzyme has a Vm value of 0. 603 mol.s^-1.g-1 , and 0, 528 mol.s^-1.g^-1 for free enzyme.
