异常黑胆质证哮喘病证结合模型的建立及评价 被引量:3

Establishment and evaluation of combining disease and syndrome model of asthma with abnormal savda syndrome

摘要 目的:建立并评价异常黑胆质证哮喘病证结合模型。方法:采用多因素复合作用建立异常黑胆质证模型,结合卵蛋白致敏激发,进一步建立异常黑胆质证哮喘病证结合模型,观察其一般状态、行为学、下丘脑-垂体-肾上腺轴(HPAA)功能、交感/副交感神经功能、气道反应性、炎症、胶原沉积等相关指标的变化。结果:异常黑胆质证组、异常黑胆质证哮喘组出现体质量下降(P<0.05),饮食水量增加(P<0.05),在旷场试验中自主活动减少(P<0.05),对糖水偏爱程度下降(P<0.05)等表现,血浆促肾上腺皮质激素(ACTH)、皮质酮(CORT)水平升高(P<0.05),环磷酸腺苷(cAMP)、cAMP/环磷酸鸟苷(cGMP)下降(P<0.05);哮喘组、异常黑胆质证哮喘组出现气道阻力升高(P<0.05),血清、肺泡灌洗液中Th1/Th2细胞因子失衡,呈Th2优势型(P<0.05),HE染色显示炎细胞浸润明显(P<0.05),PAS染色提示黏液分泌增多(P<0.05),MASSON染色提示胶原沉积增多(P<0.05),其中,以异常黑胆质证哮喘组改变更为突出。结论:采用多因素复合和卵蛋白致敏激发建立的异常黑胆质证哮喘模型一方面具有类似维吾尔医异常黑胆质证的临床表现及相关生理学指标的改变,另一方面具有气道高反应性、气道炎症、气道重塑等哮喘疾病特征,表明该模型的建立具有一定的可行性。 Objective: To establish and evaluate a model combining disease and syndrome of asthma with abnormal savda syndrome.Methods: Multiple factors were used to establish abnormal savda rat model.Then,rats were sensitized and challenged by ovalbumin and further model combining disease and syndrome of asthma with abnormal savda syndrome was established.General state,behavior,hypothalamic-pituitary-adrenal axis function,sympathetic/parasympathetic function,airway hyperresponsiveness,inflammation,collage deposition were observed.Results: Weight loss(P0.05),increased diet and water(P0.05),diminished activities in OFT test(P0.05),decreased preference for sugar(P0.05) were significantly appeared in the abnormal savda group and combining asthma with syndrome group,which was similar to the abnormal savda clinical manifestations.Furthermore,the level of CORT and ACTH in plasma were increased(P0.05),but cAMP,cAMP/cGMP were decreased(P0.05).Airway resistance was remarkably increased(P0.05) and the balance of Th1/Th2 was disturbed,which Th2 cytokines(P0.05) were dominated in the asthma group and combining disease with syndrome group.HE staining showed that inflammatory cells infiltration in the lungs(P0.05);PAS staining demonstrated mucus overproduction(P0.05) and goblet cell hyperplasia(P0.05);MASSON staining indicated emerge of collagen deposition(P0.05) in the OVA-exposed groups.Moreover,the combining disease with syndrome group was more severe than the asthma group.Conclusion: Usage of multiple factors and ovalbumin exposure could successfully establish combining disease and syndrome model of asthma with abnormal savda syndrome,and this model had either the syndrome performance of abnormal savda in Uygur medicine,or the characteristic of pathology of asthma such as airway hyperresponsiveness,airway inflammation and airway remodeling,which indicated the feasibility of this model.
