摘要
目的探讨在大鼠重度颅脑损伤后下丘脑促肾上腺皮质激素释放激素(CRH)的表达和分泌。方法应用多功能小型生物撞击机致大鼠重度颅脑损伤,采用免疫组化和PCR等技术进行检测下丘脑CRH表达的变化和规律。结果重度颅脑损伤后1h CRH表达明显增加,主要分布于室旁核和视上核,且表达呈逐渐增加的趋势,在伤后2-6h表达增高显著。结论重度颅脑损伤后早期炎症、应激反应中,下丘脑产生的CRH存在明显的增加,可能参与全身免疫炎症反应。
【Objective】To investigate increased expression and meaning of corticotropin-releasing hormone(CRH)at the hypothalamus in rats following severe head injury.【Methods】 Rats were traumatized by MuItifunctionaI BioIogicaI Impactor and then were anesthetized and sacrificed at 30 min,60 min,120 min,360 min and 720 min after severe head injury.The changes of expression and concentrations of CRH in the hypothaIamus in rats were detected by immunocytochemistry and PCR.【Resluts】One houre after severe head injury,a significant increase in CRH mRNA level was detected,and then gradually increased till 720 min after severe head injury.【Conclusion】 Expression of corticotropin-releasing hormone(CRH)at the hypothalamus in rats on the early stage following severe head injury and suggesting that CRH may play an important role through activation of some molecular pathway of immune-inflammation reactions.
