摘要
为了探讨纤维素酶对葛根中总黄酮提取的影响,试验采用单因素试验和响应曲面法对其提取工艺进行了研究,建立并分析了各主要影响因子与葛根总黄酮提取率关系的数学模型。单因素试验结果表明,液料比对葛根总黄酮提取率影响不明显、酶浓度影响极明显、酶解时间影响显著。通过RSM响应曲面法的进一步分析显示,回归方程p=0.0165〈0.01,RAdj2为0.8202和Adeq.Precision为8.671,说明所建模型与试验值的拟合度很好。葛根总黄酮的纤维素酶提取工艺参数为液料比12.1mL/g、酶浓度0.46%和酶解时间67.9min;经试验验证,在此条件下得率为0.184%,与理论计算值0.182%基本一致。
To investigate the impact of cellulase on the total flavonoids extraction from Radix Puerariae Lobatae,experiment design of single factor experiments and response surface methodology(RSM) were applied.A mathematical model was developed to analyze the relationships between the main factors and the response of the extraction yield.The results of the single factor experiments showed that solvent to material ratio was not insignificant,enzyme concentration and enzymolysis time were all very significant.The results of the response surface methodology showed that the p value 0.0165 was lower than 0.01,R_(Adj)~2 were 0.8202 and Adeq.Precision was 8.671 It indicated that the model was adequate to describe the experimental value.Based on the canonical analysis,the optimum extraction conditions were obtained as follows: solvent to material ratio 12.1 ml/g,enzyme concentration 0.46%and enzymolysis time 67.9min.Under the optimized extraction conditions,the total flavonoid extraction yield was 0.184%,which was very close to the estimated value of 0.182%by regression model.
