摘要
目的观察利心冲剂对心衰大鼠血流动力学的影响,探讨其抑制心肌细胞凋亡的分子机制。方法 50只Wistar大鼠随机分为5组:正常组、模型组、卡托普利治疗组、利心冲剂低剂量组、利心冲剂高剂量组。采取腹腔注射阿霉素(ADR,4mg/kg,每5天一次,共3次)复制大鼠心衰模型,观察心肌细胞病理学、血流动力学及凋亡相关基因Bcl-2、Bax表达的变化。结果与模型组比较,利心冲剂治疗组光镜下心肌损伤明显减轻;左室收缩压明显升高,左室舒张末压明显降低;抑制凋亡作用的Bcl-2基因表达增高、Bax/Bcl-2降低(P<0.05)。结论利心冲剂能改善心衰大鼠心功能及抑制心肌细胞凋亡,其机制可能与上调Bcl-2基因表达及降低Bax/Bcl-2比值有关。
Objective To observe the effects of Lixin granule(LXG) on hemodynamic in rats with adriamycin- induced heart fail- ure,and to investigate molecular mechanisms of apoptosis. Methods Fifty Wistar rats were randomly divided as follows:Normal con- trol group, adriamycin (ADR) group, captopril -treatment group, high and low dosage LXG groups. The pathological changes of myo- cardium,hemodynamic and relative gene of apoptosis (Bcl - 2 and Bax) were observed. Results Compared with ADR group, the extent of myocardium injury significantly reduced, the levels of left ventricular pressure (LVSP) significantly increased, while left ventricular end- diastolic pressure (I.VEDP) significantly decreased, the gene expression of Bct -2 was increased and the ratio of Bax to Bcl - 2 was decreased in LXG groups (P〈0. 05). Conclusion LXG could improve cardiac function and apoptosis of cardiomyocytes, and the mechanism might be related with up - regulating expression of Bcl - 2 gene and reducing the ratio of Bax to Bcl - 2.
