摘要
目的:探讨红景天苷对H2O2和高糖诱导人脐静脉内皮细胞(HUVECs)损伤的保护作用。方法:体外培养人脐静脉内皮细胞,分别以H2O2和高糖诱导内皮细胞损伤,分别给予10-6、10-5、10-4mol/L的红景天苷处理,MTT法测定细胞活力,检测LDH、MDA释放量和SOD活性。结果:结果显示红景天苷10-6、10-5、10-4mol/L组对正常人脐静脉内皮细胞皆无损伤作用;红景天苷10-5、10-4mol/L组对H2O2损伤模型的细胞活力有显著改善(P<0.05或P<0.01),红景天苷10-6、10-5、10-4mol/L组对高糖损伤模型的细胞活力均有显著改善(P<0.01),并且红景天苷10-5、10-4mol/L组能显著降低H2O2和高糖损伤组培养液中LDH和MDA释放量,升高细胞内SOD活性(P<0.05或P<0.01)。结论:红景天苷对H2O2和高糖诱导的人脐静脉内皮细胞损伤有明显的保护作用。
Objective:To investigate the protective effect of salilrosile on human umbilical vein endothelial cells(HUVECs) injured by H2O2 or high glucose.Methods:Human umbilical vein endothelial cells were cultured in vitro.The effect of salilrosile on nomal human umbilical vein endothelial cells was observed using MTT method,content of LDH in nutrient solution.The model of oxidative injure was induced by hydrogen peroxide(H2O2) and the model of high glucose injury was induced by high glucose,the viability of cell was measured using MTT method,LDH,MDA and SOD were measured.Results:Salilrosile10-6、10-5、10-4mol/L had no injury effect on nomal human umbilical vein endothelial cells.In the models of oxidative injury,compared with model group,salilrosile 10-5、10-4mol/L could obviously raise cell viability.In the models of high glucose injury,compared with model group,salilrosile 10-6、10-5、10-4mol/L could obviously raise cell viability.Salilrosile 10-5、10-4mol/L could lower LDH and MDA leakage,and raise intracellular SOD production in both injury models.Conclusion:Salilrosile can obviously protect cultured human umbilical vein endothelial cells from H2O2 injury and high glucose injury.
