摘要
目的观察牛磺酸联合葛根素对2型糖尿病大鼠肾脏的保护作用。方法将成模的40只Wistar大鼠随机分为糖尿病组(DM组)、单用牛磺酸组(Tau组)、单用葛根素组(Pue组)和牛磺酸联合葛根素组(Tau+Pue组)。8周后,测4组大鼠血浆葡萄糖(Glucose)、肌酐(Cre)、尿素氮(BUN)、超氧化物歧化酶(SOD)、丙二醛(MDA)、活性氧(ROS)、胰岛素(Insulin)及肾脏组织MDA、ROS及SOD,计算大鼠肾指数,光镜下观察肾脏的病理形态学变化。结果与DM组大鼠比较,Tau组、Pue组及Tau+Pue组血浆SOD活性和胰岛素水平升高,而Glucose、Cre、BUN、MDA、ROS和肾指数呈不同程度的降低,并且3个给药组大鼠肾脏组织MDA和ROS含量较DM组均有不同程度的降低,而SOD活性均有显著升高,其中Tau+Pue组这种变化更加明显;光学显微镜下显示:DM组大鼠肾小球毛细血管管腔闭塞,系膜细胞增多,基膜增厚和系膜基质大量堆积,肾小球体积增大。肾小管明显扩张,近曲小管上皮细胞肿胀、变性;牛磺酸联合葛根素处理组大鼠肾脏上述损伤性变化明显减轻。结论牛磺酸联合葛根素对2型糖尿病大鼠肾脏有保护作用,其机制可能与降低氧化应激水平有关。
Objective To investigate the protective effects of taurine combined with puerarin on kidney of rats with type 2 diabetes. Methods Forty Wistar rats with diabetes mellitus were randomly divided into diabetes mellitus group(DM),taurine treatment group(Tau), puerarin treatment group (Pue) and taurine combined with puerarin treatment group (Tau+ Pue). After 8- week treatment,the plasma levels of glucose,blood urea nitrogen(BUN), creatinine(Cre), insulin, malondialdehyde (MDA), superoxide clismutase (SOD) and reactive oxygen species (ROS) were measured in rat groups. The activity of SOD, MDA and ROS in kidney tissues and kidney index were detected respectively as well. The patho - morphological changes were observed with the light microscope. Results Compared with DM group,the activity of SOD and the content of insulin were higher, but glucose, BUN, Cre, MDA and ROS in plasma were lower in Tau group, Pue group and Tau + Pue group. Moreover the contents of MDA and ROS of kidney tissues and kidney index were decreased, but the activity of SOD increased in these three treatment groups than those in DM group. More significant changes were found in Tau + Pue group. Under the light microscope, we found the mesangial part expanded and the glomerular blood capillary ansa thickened, which separated the blood capillary ansa and crushed the Lumen of blood vessel into narrow in DM group, but Given the Tau + Pue,the morphology changes had been improved greatly. Conclusion These results suggest that taurine combined with puerarin could prevent the kidneys from in diabetic rats, which is related to partly inhibit oxidative stress.
