XBP-1稳转神经干细胞移植对PD大鼠模型的治疗作用

摘要 目的:通过测定XBP1-NSCs移植对帕金森大鼠模型黑质内单胺类递质水平的影响,研究XBP1-NSCs移植对PD的治疗作用。方法:将18只PD大鼠模型随机分为3组,每组6只大鼠。对照组侧脑室内移植入20μl的PBS,NSCs组移植空白神经干细胞悬液20μl,XBP1-NSCs组移植XBP-1基因修饰神经干细胞悬液20μl。于移植后第28天对各组大鼠进行神经功能评分,并取黑质脑组织高效液相色谱法测定单胺类递质多巴胺和3,4-二羟基苯乙酸的含量。结果:XBP1-NSCs组大鼠黑质内多巴胺和3,4-二羟基苯乙酸水平明显增加,且移植后大鼠行为学评分明显改善。结论:XBP1-NSCs移植可通过增加黑质内多巴胺和3,4-二羟基苯乙酸水平从而改善帕金森症状起到治疗目的。 Objective To study the treatment effect of XBP1-NSCs transplantation on Parkinson′s disease through detecting the influence on monoamine neurotransmitters levels in the rat model of Parkinson′s disease.Method 18 rat model of PD were divided into 3 groups randomly,and 6 rats each group.The rat in control group were transplanted 20 μl PBS through the lateral ventricle,while NSCs group were transplanted with 20 μl neural stem cells,and XBP1-NSCs group were transplanted with XBP-1 gene-modified neural stem cells.In 28 d after transplantation,neurobehavioral score of the rats was estimated respectively,dopamine and 3,4-dihydroxyphenylacetic acid content in nigra was detected by high performance liquid chromatography(HPLC).Results The dopamine and 3,4 dihydroxyphenylacetic acid contents in the transfection group was obviously increased,and the rotation speed of trasnfection group was decreased obviously.Conclusion The transplantation of XBP-1 gene-modified neural stem cells can increase dopamine and 3,4-dihydroxyphenylacetic acid level,so as to improve Parkinson′s symptoms.
